摘要
目的 :探讨非食管癌高发区病例的食管癌组织中聚合酶 β基因的突变情况。 方法 :利用RT PCR、SSCP及DNA序列分析的方法 ,对河南省非食管癌高发区 2 5例食管癌及其癌旁组织中聚合酶 β基因的变异情况进行分析。结果 :2 5例食管癌组织标本中 ,5例SSCP结果异常 ,序列分析显示 ,1例 6 13位A→T ,导致氨基酸序列 16 7位Lys→Ile ;2例 6 6 0位A→G ,导致第 183位Arg→Gly ;2例 6 70位A→G ,导致 186位Glu→Gly ;而对应的癌旁组织标本SSCP结果均无异常。结论 :非高发区食管癌组织中有 polβ突变存在。
Aim:To study DNA polymerase β gene (polβ) mutations in human esophageal cancer specimens from non high incidence area.Methods:By using RT PCR, SSCP, analysis of DNA sequence, the mutations of DNA polβgene in 25 cases of human esophageal cancer and the adjacent tissues from non high incidence area were tested.Results:Five specimens were found abnormal by SSCP, including one 613 nt A to T transvertion, which resulted in 167Lys to Ile, two 660 nt A to G transition,which resulted in 183Arg to Gly, and two 670 nt A to G transition, which resulted in 186Glu to Gly according to DNA sequence. However, there was no abnormality was found in adjacent tissues.Conclusion:There were mutations of DNA polβ gene in human esophageal cancer specimens from non high incidence area of human esophageal cancer.
