摘要
目的 :探讨宫颈癌组织中有无DNA聚合酶 β基因 (polβ)突变。 方法 :采用RT -PCR法、PCR -SSCP分析法对 12例正常宫颈组织、18例宫颈上皮内瘤样病变 (CIN)及 34例宫颈鳞癌组织进行polβ突变检测。 结果 :CIN及宫颈癌组织中存在polβ突变 ,宫颈癌组织中的突变率 (13/ 34)高于CIN(2 / 18) ,后者高于正常宫颈组织。测序结果表明宫颈癌组织中polβ 6 6 0位发生A→G突变。 结论 :宫颈癌组织中的DNA聚合酶
Aim: To explore the possibility of DNA polymerase β gene mutation in human cervical cancer tissues.Methods: The fresh removed specimen from 12 normal cervixes,18 cervical intraepithelial neoplasia (CIN),and 34 cervical cancers (CC) were collected and the mutation of DNA polymerase β gene were detected by RT-PcR, PCR-SSCP,and sequencing analysis. Results: DNA polymerase β gene mutation were found in the tissues of CIN and CC. The mutation rates in CC was 38.24%(13/34), significantly higher than that of CIN (2/18,11.11%) (P<0.05). The sequencing results showed 2 point mutations from A to G at nucleotide 660.Conclusion: The molecular mechanism of human cervical cancer may be related to the mutation of polymeraseβ gene.
