摘要
为制备高纯度鲟鱼硫酸软骨素,利用鲟鱼头骨和脊骨为原料,经过硫酸软骨素提取专用酶与碱性蛋白酶联合酶解新工艺,并进一步经季铵盐纯化、乙醇沉淀,制得硫酸软骨素纯品。硫酸软骨素提取专用酶和碱性蛋白酶提取的最佳工艺为:硫酸软骨素提取专用酶用量15mg/g,酶解温度55℃,酶解时间4h;碱性蛋白酶用量50mg/g,酶解温度60℃,酶解时间2h。季铵盐-乙醇沉淀法纯化最佳工艺为:季铵盐质量浓度1g/100mL、作用温度20℃,作用时间1.5h;乙醇沉淀体积分数为75%、溶液pH为7。该工艺制得的鲟鱼头骨、脊骨硫酸软骨素纯品的产率分别为19.5%、22.0%,纯度分别为94.80%、95.32%,蛋白含量分别为0.287%、0.267%。
Research aimed to prepare high pure production of chondroitin sulfate of sturgeon.The pure chondroitin sulfate from skull and spinal of cultured sturgeon was extracted by sequential hydrolysis with the chondroitin sulfate extraction of special enzyme and alkaline proteinase and purified by quaternary ammonium salt and ethanol.The optimum enzymolysis procedure by the chondroitin sulfate extraction of special enzyme and alkaline proteinase was as follows:15mg/g chondroitin sulfate extraction of special enzyme was added and kept at 55℃for 4hfor hydrolysis,and then 50mg/g alkaline proteinase was added and kept at 60 ℃for 2h.The optimum procedure of method of quaternary ammonium salt and ethanol precipitation was as follows:1g/100 mL quaternary ammonium salt solution was added and kept at 20℃for 1.5hand ethanol solution adjusted to pH 7was 75%.Under these conditions,the yield of the pure products of chondroitin sulfate from skull and spinal was 19.5% and 22.0%,the purity was 94.80% and95.32%,and the protein level was 0.287%and 0.267%,respectively.
