摘要 利用Box-behnken响应面设计,优化乙酸提取法提取蚕蛹抗菌肽工艺。选取乙酸浓度、料液比、提取时间3个对提取效果有显著影响的因素作三元二次回归模型,以提取物抑制大肠杆菌的抑菌效果作为评价标准。确定最佳提取工艺条件为乙酸体积分数5.24%,料液比1∶2.31,提取时间8.32 h时,对大肠杆菌的抑菌圈直径预期可达1.903 22 cm,实际验证1.887 00 cm,达到回归模型预测的99.14%,说明响应面法优化蚕蛹抗菌肽的提取可行。 By the Box-behnken response surface design, acetate extraction method silkworm chrysalis antimicrobial peptides process is optimized. The ternary quadratic regression model is studied with the acetic acid concentration, solid-liquid ratio, extraction time as effect factors, and extract inhibiting the antibacterial effect of E. coli as an evaluation criterion. The optimum extraction conditions are determined as follows: the acetic acid concentration of 5.24%, the solid to liquid ratio of 1∶2.31, extraction time of 8.32 h, and the E. coli inhibition zone diameter expects up to 1.903 22 cm, actual validation 1.887 00 cm, to 99.14% of the regression model to predict, indicating that the response surface method to optimize the silkworm chrysalis antimicrobial peptide extraction is feasible.
