摘要
建立银杏叶中聚戊烯醇提取物样品的水解及HPLC分析方法 以石油醚(60~90℃)为溶剂,超声萃取银杏叶中聚戊烯醇类化合物,室温下(25℃)提取物用5%的NaOH甲醇溶液水解2.5h,再经硅胶柱层析分离纯化聚戊烯醇化合物后进行HPLC分析 结果表明:聚戊烯醇乙酸酯可完全水解成聚戊烯醇,水解中聚戊烯醇类化合物结构稳定 经HPLC分析,银杏叶中聚戊烯醇化合物含量为1.90%,RSD=1.71%(n=5) 聚戊烯醇进样量在0.26~5.2μg范围内与峰面积有良好的线性关系 该方法简单、稳定。
A method was established for the hydrolysis and HPLC determination of polyprenols in ginkgo biloba leaves. The polyprenols in ginkgo biloba leaves were ultrasoundextracted three times with petroleum ether(60~90℃). Prior to HPLC analysis, the extract was hydrolyzed with alkali methanol solution for 2.5h at room temperature(25℃) and then purified by silica gel column chromatography. The results showed that the structures of polyprenols were not destroyed and the polyprenyl acetates could be converted into polyprenyl alcohols entirely in hydrolysis process. The content of polyprenols in ginkgo leaves was determined to be 1.90% by HPLC method with a RSD of 1.71%(n=5) and the calibration curve was linear in the range of 0.26~5.2μg. This method is simple, stable and can be used for the analysis of polyprenols in ginkgo biloba leaves.
