摘要
目的 探讨去卵巢 (ovariectomy ,OVX)、去松果体 (pinealectomy ,PX)对大鼠齿状回 (Dentategyrus ,DG) β淀粉样蛋白 (β amy loidprotein ,Aβ)损伤区突触泡膜素 (synaptophysin ,SYN)表达的影响。 方法 将大鼠随机分为假手术、生理盐水 (NS)对照、正常 +Aβ、假OVX +Aβ、OVX +Aβ、假PX +Aβ、PX +Aβ、假OVX +假PX +Aβ、OVX +PX +Aβ 9组。术后 60d,将Aβ注入DG背侧带分子层 ,7d后以SYN作标记 ,用SABC法检测Aβ损伤区DG背侧带分子层的SYNPu值。 结果 假手术组与NS对照组无DG神经元损害、DG背侧带各分子层SYNPu值无显著差别 ,合并为B1组 ,假手术 +Aβ各组与正常 +Aβ组的DG神经元损害、DG背侧带各分子层SYNPu值无显著差别 ,合并为B2组。各组Aβ直接接触的DG分子层SYNPu值均明显减低 ,组间无明显区别。Aβ注射点以外 ,各组间DG背侧带内外分子层SYNPU值有明显区别 :OVX +PX +Aβ组 <PX +Aβ组 <OVX +Aβ组 <B1组 <B2组 ;各组间DG背侧带中分子层无显著区别。结论 Aβ 40对DG分子层突触有直接的损害作用。在Aβ注射点以外区域 ,OVX、PX使Aβ的DG颗粒细胞损害诱导的其背侧带内外分子层的代偿性神经生长消失 ,PX比OVX危害大 。
Objectives To investigate the effects of OVX and PX on expression of SYN in lesion areas by Aβ of DG of rats. Methods The rats were randomly divided into 9 groups: sham operation , NS control , normal+ Aβ, Sham OVX+Aβ, OVX+Aβ, Sham PX+Aβ, PX+Aβ, Sham OVX+ Sham PX+Aβ, OVX+PX+Aβ. 60d after surgery, Aβ1 40 was injected into the molecular layers of dorsal blade of DG. 7d After surgery , SYN Pu values(SYN Pu) of lesion areas by Aβ of molecular layers of DG dorsal blade were detected by SABC method with SYN as a marker. Results Since there was no DG lesion in sham operated group and NS control group , and there were no significant difference in SYN Pu value of the molecular layers of dorsal blade of DG, so they were pool into group B1 . There were no significant difference about neurons lesion of hippocampus and DG and SYN Pu value of the molecular layers of dorsal blade of DG among all sham operated+Aβ groups and normal+Aβ group , so they were combined to group B2 . The SYN Pu values of molecular layers of DG contacting directly with Aβ1 40 in all Aβ1 40 injection groups were significantly declined , but no significant difference among all groups .There were significant difference in SYN Pu values of inner and outer molecular layers of dorsal blade of DG suffering granule cells damage by Aβ1 40 except the site of Aβ1 40 injection among all groups , the SYN Pu values were in following gradation : OVX+PX+ Aβ group <PX+ Aβ group < OVX+Aβ group <group B1<group B2,but there were no significant difference in SYN Pu values of middle molecular layers among all groups .Conclusions Aβ1 40 had direct neurotoxicity on synapses of DG molecular layers in vivo . OVX or PX make compensatory synaptic sprouting of the molecular layers in dorsal blade of DG induced by neurons lesion of DG by Aβ1 40 disappear, and the effect caused by PX was more serious than by OVX , moreover collective effect was even much more serious.
