摘要
目的:探讨去卵巢、去松果体对海马、齿状回细胞凋亡的影响,了解雌激素、褪黑素缺乏与阿尔茨海默病发病及进展的关系。方法:将大鼠随机分为正常、假去卵巢、去卵巢、假去松果体、去松果体、假去卵巢+假去松果体、去卵巢+去松果体7组。术后90d,用苏木精-伊红、TUNEL染色观察去卵巢、去松果体对海马细胞凋亡的影响。结果:假手术各组与正常组在各项检测指标上无明显差异,合并为A组,A组在海马、齿状回偶有细胞凋亡但部位不恒定,去卵巢、去松果体、去卵巢+去松果体各组均出现显著增多的细胞凋亡,并且去松果体+去卵巢组(17.75±3.43)个>去松果体组(14.27±1.35)个>去卵巢组(10.01±2.22)个>A组(6.09±2.21)个,差异有非常显著性意义(P<0.01);A组犤(20.92±5.77)个犦、去卵巢组犤(23.04±2.31)个犦、去松果体组犤(22.25±5.01)个犦、去松果体+去卵巢组犤(23.27±4.78)个犦海马、齿状回无明显胶质细胞增生(P>0.05)。结论:去卵巢、去松果体可诱发海马、齿状回细胞凋亡显著增多,且去松果体比去卵巢影响大,去卵巢+去松果体影响更大;这种影响与胶质细胞无关。
AIM:To investigate the influence of ovariectomy(OVX) and pinealectomy(PX) on the apoptosis of hippocampus and dentate gyrus(DG) in rats,and study the relation of the deficiencies of estrogen and melatonin with the occurrence and development of Alzheimer disease. METHODS:The rats were randomly divided into 7 groups:normal group,sham OVX group,OVX group,sham PX group,PX group,sham OVX+sham PX group and OVX+PX group.Ninety days after surgery,effect of OVX and PX on apoptosis in hippocampus and DG of rats were observed by HE and TUNEL methods. RESULTS:The normal group and all sham operated groups were combined to group A for having no significant discrepancy in all detected indexes,apoptosis appeared randomly without consistent areas in hippocampus and DG in group A.Compared with group A,the number of apoptosis in hippocampus and DG was increased significantly in the OVX,PX and OVX+PX groups,and PX +OVX group(17.75±3.43) >PX group(14.27±1.35) >OVX group (10.01±2.22) >group A(6.09±2.21),there were very significant differences(P< 0.01).There were no significant differences in the glial cells hyperplasia in hippocampus and DG among group A(20.92±5.77),OVX group(23.04±2.31),PX group(22.25±5.01) and PX+OVX group(23.27±4.78)(P >0.05). CONCLUSION:OVX or PX can induce the significant increase of apoptosis in hippocampus and DG,and the effect of PX is greater than that of OVX,moreover that of OVX+PX is much greater,which has no relation with glial cells.
