氧氟沙星喷雾剂体外杀菌实验方法的改进 被引量:1

摘要 目的 采用微孔滤膜过滤和比浊法测试不同浓度的氧氟沙星喷雾剂稀释液的体外杀菌效果 ,并与菌落计数法比较 ,考察该法的可行性。方法 将自备氧氟沙星喷雾剂分别稀释 10、10 0倍 ,分别作用于供试绿脓杆菌菌液 15、30min ,然后用微孔滤膜过滤比浊法测试其杀菌效果 ,并设置对照组进行比较。在扫描电镜下观察受作用后的菌态的变化。同时用菌落计数法进行比较。结果 微孔滤膜可拦截供试菌 ,蒸馏水 (10 0ml× 3)冲洗氧氟沙星作用后的带菌滤膜可视为喷雾剂菌液已无残余。比浊法与菌落计数法相比 ,可得到等同的实验结果 ,并且更简便、快捷。结论 微孔滤膜过滤和比浊法二者相结合是一种方便、省时。 OBJECTIVE To test the disinfecting effect of Ofloxacin aerosol in different diluted concentrations by turbidimetry after filtrating through micro-aperture filter membrane and evaluate the feasibility of the method comparing with colony-count method.METHODS The self-provided ofloxacin aerosol were diluted 10 times,100 times and reacted on prepared Bacillus pyocyaneus for 15 min,and 30 min respectively.Then filtrated through micro-aperture filter membrane and tested the disinfectant effect by turbidimetry compared with the control groups.Also the reactant was colony-count method was adopted as a comparison. The morphological changes of bacteria after reaction were observed through a scanning electron microscope.RESULTS The micro-aperture filter membrane was capable of holding the tested bacteria.The filter membrane with bacteria could be regarded that there were no ofloxacin left after washing by distilled water(100 ml×3).Compared with colony-count method,the method of turbidimetry is much more esaier and quicker and also can get effective results.CONCLUSION The combination of two methods is a convenient,rapid and feasible exoteric disinfectant experimental method.
