IL-10 regulates macrophage polarization

摘要 目的研究白细胞介素10(IL-10)在骨髓单个核细胞向巨噬细胞分化以及在巨噬细胞极化中的作用。方法分离野生型及IL-10表达缺陷小鼠的骨髓单个核细胞,巨噬细胞集落刺激因子(M-CSF)诱导细胞分化为巨噬细胞。流式细胞仪检测细胞的增殖、分化、凋亡以及脂多糖(LPS)刺激后巨噬细胞的极化;定量PCR检测细胞周期调控相关分子及炎性相关分子的表达。结果骨髓单个核细胞诱导5~7 d,流式细胞仪检测结果显示,IL-10的表达缺陷不影响细胞向巨噬细胞的分化。相比野生型,IL-10缺陷细胞数量虽略有增加,但差异不显著。流式细胞仪检测结果及定量PCR结果进一步证明,IL-10表达缺陷不影响细胞增殖和凋亡。IL-10缺陷细胞分化7 d后CD86的表达显著高于野生型,LPS刺激后CD86表达均升高,但IL-10缺陷巨噬细胞的表达显著高于野生型。LPS刺激6h,IL-10缺陷巨噬细胞中诱导型一氧化氮合酶(i NOS)和IL-18的表达水平显著高于野生型巨噬细胞。结论 IL-10不影响单个核细胞的增殖分化,但参与调控巨噬细胞的极化。 Objective To explore the role of IL-10 in the development of bone marrow derived macrophages and their polarization. Methods Mononuclear cells( BMCs) were isolated from the bone marrow of WT and IL-10 deficient mice and treated with M-CSF for macrophage differentiation. Flow cytometry assay was used to examine the proliferation,differentiation,apoptosis and polarization of macrophages. Quantitative PCR assay was performed to analyze the expression of related molecules. Results The mRNA level of IL-10 in IL-10 deficient cells was significantly lower than that of WT counterparts. Flow cytometry data and quantitiative PCR data indicated that the differentiation of BMCs toward macrophages and their proliferation were similar in WT and IL-10 deficient mice. Comparable cell death was also detected in IL-10 deficient cells,whether M-CSF was deprived or not. In the LPS-condition medium,however,IL-10 tm1/tm1 macrophages showed a higher expression of surface marker CD86 and more pro-inflammatory factors IL-18 and i NOS production.Conclusion These data indicate that IL-10 is involved in macrophage polarization.
