摘要
目的 :比较离心管、脱钙松质骨和微囊三种方式立体培养软骨细胞的特点。方法 :采用海藻酸钠 -多聚赖氨酸 -海藻酸钠微囊将永生化下颌骨髁突软骨细胞 (immortalizedmandibularcondylarchondrocyte ,IMCC)包裹 ,同时将IMCC接种于离心管和脱钙的松质骨上 ,定期观察细胞的形态及生长情况 ,然后将立体生长的软骨细胞进行组织学观察。结果 :IMCC在微囊聚集成团 ,形成一定的组织样结构。细胞附着于松质骨的表面 ,也可以形成薄层样结构。生长在离心管内的细胞形态较差 ,形成的立体组织结构不良。结论 :采用微囊包裹软骨细胞的方法可以在体外短期内大量、快速、立体培养软骨细胞 。
Objective:To compare three methods for chondrocytes cultre in three-dimension.Methods:Alginate-polylysine-alginate(APA) microcapsule technique was used to encapsulate immortalized mandibular condylar chondrocyte (IMM).Also IMCC cells were seeded in centrifugal tubes and decalcified bone.The proliferation and morphology of cells were observed,then histological assay was performed to observed cultured IMCC cells in three-dimension.Results:IMCCs proliferated in culsters and formed tissue-like construction within microcapsules.IMCCs seeded to the surface of decalcified bone and also formed membrane-like construction.IMCCs seeded in centrifugal tubes showed a bad cellular shape and construction.Conclusions:Microcapsule technique can be used to culture chondrocytes rapidly in three-dimension in a large scale,which is suitable to research on cartilage tissue engineering for small and middle laboratory.
