摘要
目的 :对小鼠血清中阿霉素的含量进行了测定 ,以便对其进行血药浓度监测及药物动力学研究。方法 :建立了以α 萘酚为内标的反相高效液相色谱法 (HPLC) ,并对用药后小鼠血清中的阿霉素的含量进行了测定。结果 :血清样品用氯仿 甲醇 ( 4∶1)混合液 1ml提取后进样。色谱柱 :Nova pakC1 8柱 ( 3 .9mm× 15 0mm) ,流动相 :甲醇 乙腈 0 .0 1mol·L- 1 NH4H2 PO4 冰HAc( 4 0∶10∶5 0∶0 .6 ) ,检测波长 :2 33nm。线性范围 :0 .1~ 10 .0 μg·ml- 1 ,r =0 .9994。结论 :该法快速、简便 ,内标价廉、易得 ,适于临床药物浓度监测及药物动力学研究。
Objective:A HPLC method for the determination of doxorubicin in mice serum was reported to study the pharmacokinetics of Dox. Methods:α-naphthol was used as the internal standard. The method has been applied to study the pharmacokinetics of doxorubicin after intravenous injection in mice. Results:The Dox was extracted from serum by trichloromethane-methanol(4∶1). A solution of methanol-acetonitrile-0.01mol·L -1NH4H2PO4-acetic acid (40∶10∶50∶0.6)was used as the mobile phase.The detection of wavelengths was set at 233nm. The calibration curve was linear in the range from 0.1 μg·ml -1 to 10.0 μg·ml -1 with r=0.9994. Conclusions: The internal standard is convenient to acquire. The method is rapid,simple and easy to use in TDM and the study of the pharmacokinetics of Dox.
