石菖蒲挥发油中β-细辛醚在小鼠体内的吸收、分布和排泄研究 被引量:4

摘要 目的 研究石菖蒲挥发油中β-细辛醚在小鼠体内的药动学。方法 采用气相色谱法测定β-细辛醚的血药浓度,色谱柱:HP-35毛细管气相色谱柱;程序升温:起始温度为30℃,6℃·min-1升至220℃,保持5 min;进样口温度:250℃;检测器温度:320℃;进样量:2.0μL;无分流进样;溶剂:甲醇;载气:氦气;柱头压:5.5×104 Pa;内标物:α-萘酚。结果 β-细辛醚与内标物的峰面积之比(y)与浓度(c)之间的回归方程为:y=0.018 89c+1.755 × 10-3(n=6,r=0.999 9)。线性范围:0.04-40.0μg·mL-1。小鼠全血中最低检测浓度为:0.04μg·mL-1。结论 小鼠灌胃石菖蒲挥发油后,β-细辛醚在体内的药-时过程为线性动力学过程,符合一级吸收二室模型,t1/2(α)为6.5 min,t1/2(β)为93.6 min。 OBJECTIVE: To study the pharmacokinetics of β-asarone in volatile oil from Acorus tatarinowii Schott, in mice. METHODS: A capillary GC method was applied to determine the concentration of β-asarone in blood. HP-35 column was used as the analysis column. Helium was used as carrier gas. The oven temperature was started at 30°C, then raised to 220°C by a step of 6°C&middotmin-1 and stayed for 5 minutes. The injector and detector temperatures were 250°C and 320°C respectively. Injection volume was 2.0 μL with splitless inject mode. Methanol was used as the solvent and α-naphthol as the internal standard. RESULTS: A capillary GC method was established for the determination of β-asarone in mouse blood. The linear equation Y = 0.018 89c + 1.755 × 10-3 (n = 6, r = 0.999 9) was obtained with in the range of 0.04 ∼ 40 μg&middotmL -1. The limit of quantification was 0.04 μg&middotmL-1 and the recovery of β-asarone in blood was 95.3% (RSD = 3.92%). CONCLUSION: After oral administration of the volatile oil in mice, the pharmacokinetic process of β-asarone accorded with 2-compartment model with 1st order absorption, tmax, t1/2α and t 1/2β were 10,6.5 and 93.6 min respectively. The absorption and elimination of β-Asarone were rapid, β-asarone was detected in tissues at 5 min following administration. The drug concentrations in most of the tissues achieved a high level within 10 min. The concentration in the small intestine was the highest. Those in the kidney, heart, blood, liver, lung, brain, spleen and muscle followed in turn. The drug concentrations in all these tissues decreased more than 90% within 7 h.
