海参硫酸多糖的高温高压降解工艺及其降解机制 被引量:13

摘要 目的分析海参硫酸多糖的高温高压降解工艺和降解产物的结构,探究海参硫酸多糖的高温高压降解的机理。方法采用聚丙烯酰胺凝胶电泳法、分子排阻凝胶色谱法、薄层层析法、离子高效液相色谱法和PMP柱前衍生-高效液相色谱法分析多糖的分子量、硫酸根含量和单糖组成的变化,并利用液相色谱-质谱联用技术分析降解产物中硫酸寡糖的结构特征。结果当pH>7时,海参硫酸多糖在高温高压条件下未见明显降解。添加醋酸使溶液的pH降至6以下,海参硫酸多糖在100~121℃加热后,pH越低、温度越高、加热时间越长,分子量降低越明显。海参硫酸多糖的硫酸根和单糖组成在降解后变化很小,降解产物中存在一系列奇数和偶数的硫酸岩藻寡糖和硫酸软骨素寡糖,说明高温高压降解为无规断裂型和解聚断裂混合型。相同条件下,岩藻聚糖硫酸酯比硫酸软骨素更易被降解。结论本研究建立了高温高压法高效降解海参硫酸多糖的技术,该方法是1种回收率高、易控制、绿色环保的低分子量海参硫酸多糖的制备方法。 Objective To explore the mechanism of hydrothermal degradation of sulfated polysaccharides from sea cucumber and analyze the degradation process and structures of degradation products.Methods Polyacrylamide gel electrophoresis,high performance molecular exclusion gel chromatography,thin layer chromatography,ionic liquid chromatography and PMP precolumn derivatization-high performance liquid chromatography were used to analyze the change of molecular weight,sulfate content,and monosaccharide composition of sulfated polysaccharides during degradation.The structures of sulfated oligosaccharides in the degradation products were analyzed by HILIC-MS.Results When pH>7,there was no obvious degradation of sulfated polysaccharides after hydrothermal treatment.When pH was 6 or less with addition of acetic acid,the molecular weight of sulfated polysaccharides from sea cucumber decreased significantly heated at 100~121℃.The lower the pH,the higher the temperature,and the longer the heating time,the lower the molecular weight was.The monosaccharide composition and sulfate content of sulfated polysaccharides did not change much during hydrothermal degradation.A series of odd and even number of sulfated fucan and chondroitin sulfate oligosaccharides were produced after hydrothermal degradation analyzed by hydrophilic liquid chromatography-mass spectrometry,indicating that sulfated polysaccharides were degraded by a mixed type of random fracture and depolymerization fracture during hydrothermal degradation.Fucoidan was more easily degraded than chondroitin sulfate under the same condition.Conclusion A hydrothermal degradation process in mild acid situation to obtain low molecular weight sulfated polysaccharides was established.This method has advantages of high recovery rate,easy control and environmental protection.
