摘要
目的:观察白子菜总黄酮对脑缺血模型大鼠预处理的保护作用,探讨白子菜总黄酮提高脑缺血模型大鼠耐受作用机制。方法:80只SPF级Wistar大鼠随机分为假手术组,模型组,阳性对照组(0.02 g·kg^(-1)),白子菜总黄酮大剂量组、小剂量组(200 g·kg^(-1)、100 g·kg^(-1))。采用阻断双颈血流10 min,脑缺血预处理,恢复血流灌注2 h后,连续灌胃给药18 d,再次阻断双颈血流30 min,再灌注24 h后,建立脑缺血模型大鼠耐受动物模型。HE染色,观察大脑病理变化;酶联免疫法(ELISA)检测脑组织匀浆液白细胞介素-17(interleukin-17,IL-17)、IL-10、Bcl-2、Bax、Caspase-3的水平。结果:与模型组比较,白子菜总黄酮大剂量、小剂量组IL-17、Bcl-2、Caspase-3水平显著降低(P<0.01,P<0.05);白子菜总黄酮大剂量组、小剂量组IL-10、Bax水平显著升高(P<0.01);白子菜总黄酮能有效改善脑缺血再次出血造成的脑组织损伤,降低神经元细胞损伤。结论:白子菜总黄酮能提高脑缺血耐受作用,其作用机制可能与白子菜总黄酮降低细胞凋亡有关。
Objective:To observe the protective effect of total flavonoids of Gynura divaricata on the preconditioning of rats with cerebral ischemia,and to explore the mechanism of tolerance of total flavonoids of Gynura divaricata.Methods:Eighty SPF Wistar rats were randomly divided into sham operation group,model group,positive control group(0.02 g·kg-1),high dose group of white broccoli total flavonoids,and low dose group(200 g·kg-1),(100 g·kg-1).After blocking the blood flow of the double neck for 10 min,the cerebral ischemic preconditioning,and the blood perfusion for 2 hours,the rats were administered with continuous intragastric administration for 18 days,the blood flow of the double neck was blocked again for 30 minutes,and the rats with cerebral ischemia tolerant animal models were established after 24 hours of reperfusion.HE staining was used to observe the pathological changes of brain;enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of interleukin-17(IL-17),IL-10,Bcl-2,Bax and Caspase-3 in brain homogenate.Results:Compared with the model group,the levels of IL-17,Bcl-2 and Caspase-3 in the high-dose and low-dose groups of the total flavonoids of Radix Paeoniae Alba were significantly decreased(P<0.01,P<0.05).The levels of IL-10 and Bax in the dose group were significantly increased(P<0.01).The total flavonoids of Radix Paeoniae Alba could effectively improve brain tissue damage caused by cerebral ischemia and rebleed,and reduce neuronal cell damage.Conclusion:Total flavonoids of Gynura divaricata can improve cerebral ischemic tolerance,and its mechanism may be related to the decrease of apoptosis by total flavonoids of Chrysago chinensis.
