葛根总黄酮通过下调NF-κB信号通路改善力竭运动小鼠的心肌损伤 被引量:2

Total Flavonoids from PUERARIAE LOBATAE RADIX Alleviate Exhaustive Exercise-Caused Myocardial Injury in Mice by Down-Regulating NF-κB Signaling Pathway

摘要 目的:研究葛根总黄酮对力竭运动小鼠心肌损伤的保护作用及其机制。方法:健康成年雄性SPF级BALB/c小鼠48只随机分为:正常对照组、模型对照组、地奥心血康100 mg/kg组、葛根总黄酮25、50、100 mg/kg组。正常对照组和模型对照组给予生理盐水灌胃,其余各组给予相应药物。实验组小鼠进行7 w的无负重游泳训练,于第50 d上午进行游泳训练直至力竭。观察心肌损伤情况,检测血清中肌酸激酶同工酶(CK-MB)、白细胞介素-1β(IL-1β)、IL-6、肿瘤坏死因子-α(TNF-α)的含量,检测心肌组织丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH)的含量、超氧化物歧化酶(SOD)活力;Western blot法检测心肌中相关转录因子(IKKα、p-IKKα、IκBα和p-IκBα)的蛋白表达。结果:与正常对照组相比,模型对照组血清CK-MB、IL-1β和TNF-α、心肌中MDA含量均显著升高(P<0.01),GSH含量、SOD活力显著降低(P<0.01);与模型对照组相比,葛根总黄酮50 mg/kg组血清CK-MB、IL-1β和TNF-α、心肌MDA含量显著降低(P<0.01),心肌GSH含量和SOD活力明显升高(P<0.05);心肌中p-IKKα在葛根总黄酮干预后表达下调,p-IκBα的表达上调。结论:葛根总黄酮可提高心肌组织抗氧化酶活性和抗炎能力,对力竭运动引起的心肌损伤有良好的保护作用,其机制可能与NF-κB信号通路有关。 Objective:To study the protective effect and mechanism of total flavonoids from PUERARIAE LOBATAE RADIX on exhaustive exercise-caused myocardial injury in mice. Methods:Forty-eight healthy SPF-grade adult male BALB/c mice were randomly assigned into six groups(n=8):a normal control group, a model control group, a Diaoxinxuekang(100 mg/kg) group, and low-,medium-,and high-dose(25,50,and 100 mg/kg) groups of total flavonoids from PUERARIAE LOBATAE RADIX. The normal control and model control group were administrated with normal saline by gavage, and the other groups with corresponding drugs. The mice in other groups except the normal control group were subjected to 7 weeks of weight-free swimming training. In the morning of day 50,swimming training was carried out until exhaustion. The myocardial injury was observed, and the serum levels of creatine kinase-MB(CK-MB),interleukin-1 beta(IL-1β),IL-6,and tumor necrosis factor-alpha(TNF-α) were determined. The content of malondialdehyde(MDA) and the activities of superoxide dismutase(SOD) and glutathione peroxidase(GSH) in the myocardial tissue were measured. Western blotting was employed to examine the expression levels of IκB kinase α(IKKα),phosphorylated IKKα(p-IKKα),inhibitor of nuclear factor kappa B alpha(IκBα),and p-IκBα in the myocardium. Results:Compared with the normal control group, the modeling elevated the serum levels of CK-MB,IL-1β,and TNF-α,increased the MDA content in the myocardium(P<0.01),and decreased the activities of GSH and SOD(P<0.01). Compared with the model control, 50 mg/kg total flavonoids from PUERARIAE LOBATAE RADIX lowered the serum levels of CK-MB,IL-1β,and TNF-α,reduced the MDA content in the myocardium(P<0.01),and increased the activities of GSH and SOD(P<0.01). Moreover, the total flavonoids down-regulated the expression level of p-IKKα and up-regulated that of p-IκBα. Conclusion:The total flavonoids from PUERARIAE LOBATAE RADIX can improve the antioxidase activity and anti-inflammatory capacity of the myocardial tissu
