摘要
目的:探讨白鲜皮提取物的抗过敏作用,分析其作用机制。方法:采用C48/80处理大鼠肥大细胞RBL-2H3,刺激RBL-2H3细胞脱颗粒,释放组胺及β-氨基己糖苷酶,模拟人类过敏反应。用不同浓度的白鲜皮提取物处理大鼠肥大细胞RBL-2H3,观察白鲜皮提取物对大鼠肥大细胞RBL-2H3脱颗粒释放组织胺及β-氨基己糖苷酶的影响;采用伊文思兰染色的方法,观察白鲜皮提取物对组织胺引起的毛细血管通透性的影响。结果:发现白鲜皮提取物能够剂量依赖性的抑制C48/80诱导的大鼠肥大细胞RBL-2H3脱颗粒,减少组织胺及β-氨基己糖苷酶的释放,并能够降低小鼠毛细血管通透性。结论:白鲜皮提取物能够缓解过敏反应引起的炎症而用于过敏反应,其作用机制可能是通过抑制肥大细胞功能而降低毛细血管通透性。
Objective: To explore the anti-allergic effect of the extract of densefruit pittany root-bark and analyze its mechanism.Methods: Rat mast cell RBL-2 H3 was treated with C48/80 to stimulate RBL-2 H3 cell degranulation,release histamine and β-aminohexosidase,and simulate human allergic reaction. Rat mast cells RBL-2 H3 were treated with different concentrations of extract of densefruit pittany root-bark. The effects of extract of densefruit pittany root-bark on the release of histamine and β-aminohexosidase from the degranulation of rat mast cells RBL-2 H3 were observed. The effect of extract of densefruit pittany root-bark on capillary permeability induced by histamine was observed by Evans blue staining. Results: It was found that the extract of densefruit pittany root-bark could inhibit the degranulation of RBL-2 H3 induced by C48/80 in a dose-dependent manner,reduce the release of histamine and β-aminohexosidase,and decrease the capillary permeability in mice. Conclusion: The extract of densefruit pittany rootbark can alleviate the inflammation caused by allergic reaction and be used for allergic reaction. Its mechanism may be to reduce capillary permeability by inhibiting mast cell function.
