摘要
目的:观察滋阴清热方对狼疮鼠脑组织细胞因子白细胞介素-1β(interleukin-1β,IL-1β)和补体裂解产物C5a表达的影响。方法:SPF级B6.MRL-Faslpr/Jnju 6周龄雌性小鼠20只随机平均分为模型组和中药组;SPF级C57BL/6JN 6周龄雌性小鼠10只为正常组。中药组按滋阴清热方20 g·(kg·d)^(-1)剂量灌胃(人等效剂量的2倍);模型组给予等容积蒸馏水灌胃;正常组给予等容积蒸馏水灌胃。干预疗程6周。6周后取脑组织,应用免疫组织化学、荧光定量PCR检测IL-1β、C5a在脑膜的表达。结果:滋阴清热方干预后,中药组IL-1β和C5a免疫组织化学染色较模型组变浅,阳性位点减少。荧光定量PCR结果显示:①IL-1β在脑膜的表达量:模型组表达量高于中药组,但差异无统计学意义(P=0.535);模型组和中药组均高于正常组,差异均有统计学意义(P=0.00,P=0.01)。②C5a在脑膜的表达量:模型组高于中药组和正常组,差异均有统计学意义(P=0.00,P=0.00);中药组高于正常组,差异有统计学意义(P=0.001)。结论:滋阴清热方可降低IL-1β和C5a在狼疮鼠脑膜的表达,可改善狼疮鼠脑组织免疫性血管炎,保护血脑屏障的完整性,进而缓解狼疮脑病的进程。
Objective: To observe the effect of nourishing yin and clearing heat prescription on the expression of interleukin-1β( IL-1β) and complement cleavage product C5 a in brain tissue of lupus rats. Methods: 20 SPF-class B6. MRL-Faslpr/Jnju 6-week-old female mice were randomly divided into model group and Chinese medicine group;10 SPF-class C57 BL/6 JN 6-week-old female mice were normal group. The Chinese medicine group was intragastrically administered with a dose of 20 g·( kg·d)-1 of nourishing yin and clearing heat prescription( two times the equivalent dose of human);the model group was given equal volume of distilled water for gavage;the normal group was given equal volume of distilled water for gavage,with intervention treatment for 6 weeks. Brain tissue was taken after 6 weeks,and the expression of IL-1β and C5 a in the meninges was detected by immunohistochemistry and quantitative fluorescence PCR. Results: After the intervention of nourishing yin and clearing heat prescription,the immunohistochemical staining of IL-1β and C5 a in the Chinese medicine group was lighter than that in the model group,and the positive sites were reduced. The results of quantitative fluorescence PCR showed that the expression of IL-1β in the meninges was higher in the model group than in the Chinese medicine group,but the difference was not statistically significant( P = 0. 535).Both the model group and the Chinese medicine group were higher than the normal group,and the difference was statistically significant( P = 0. 00,P = 0. 01).(2)The expression of C5 a in the meninges: the model group was higher than the Chinese medicine group and the normal group,the difference was statistically significant( P = 0. 00,P = 0. 00);the Chinese medicine group was higher than the normal group,the difference was statistically significant( P = 0. 001). Conclusion: Nourishing yin and clearing heat prescription can reduce the expression of IL-1β and C5 a in the meninges of lupus rats,improve the immune vasculitis in the brain tissue of lupus
