摘要
目的研究C5 a反义肽(NH2-EARLQRVFLYVNPS-COOH)对盲肠结扎穿孔(cecal ligation and puncture,CLP)致脓毒症小鼠发生肺损伤的保护性作用。方法实验组小鼠CLP后即经尾静脉注射反义肽R4 1、2、4、8 mg/kg,浓度0.5mg/m l),观察不同剂量R4对小鼠96 h生存率的影响,并对肺组织进行病理观察,检测肺髓过氧化物酶(MPO)活性,检测动脉血氧分压、肺湿干比。结果相较于对照组,治疗组小鼠在R4使用浓度2 mg/kg时96 h生存率明显提高(P<0.05),动脉氧分压升高(P<0.01)、MPO和肺湿/干比降低(P<0.01)、病理改变减轻。结论反义肽R4在小鼠体内有良好的拮抗C5 a作用,对脓毒症小鼠肺的急性损伤有保护作用。
Objective To evaluate the protective effects of the antisense peptide of C5a ( NH2-EARLQRVFLYVNPS-COOH) on lung injury of mouse sepsis Mice were subjected to cecal ligation and puncture model by cecal ligation and puncture (CLP). Methods (CLP). Forty mice received 0.5 mg/ml antisense peptide of C5a at the dose of 1, 2, 4, 8 mg/kg through the tail vein after CLP, line as control and the survival rate was observed. The pathological c with ten mice receiving physiological sahange of the lung and the arterial partial pressure of oxygen, lung wet weight/dry weight and myeloperoxidase (MPO) were determined in normal controis, sham operation mice, CLP mice and CLP mice recceiving antisense peptide at dose of 4 mg/kg (n = 10 in each group). Results The mice receiving antisense peptide at dose of over 2 mg/kg had higher 96-hour survival rate (P 〈 0.05 ), higher arterial partial pressure of oxygen ( P 〈 0.01 ), the decreased activity of MPO and lung wet weight/dry weight ( P 〈 0.01 ), less pathological changes than the controls. Conclusion Our study suggests that the antisense peptide of C5a has an antagonistic effect against C5a in vivo and a possible new therapeutic drug category for sepsis.
