摘要
目的:研究白杨素(chrysin,ChR)抑制肺癌A549细胞系球形成细胞自我更新作用。方法:不同浓度ChR处理体外培养A549细胞系球形成细胞。肿瘤球形成法测定自我更新。Western blot分析CK2α和Gli1蛋白表达。pcDNA3.1-CK2α转染探讨作用机制。结果:ChR(10、20和40μM)处理24小时显著降低A549细胞系肿瘤球形成率(P<0.05),并伴随着CK2α和Gli1蛋白表达下调。与pcDNA3.1-LacZ转染细胞相比,pcDNA3.1-CK2α转染A549细胞系球形成细胞CK2α和Gli1蛋白表达显著上调,并增高肿瘤球形成率和减弱ChR的作用。结论:下调CK2α和Gli1表达参与ChR抑制A549细胞系球形成细胞自我更新作用。
Objective To investigate the effect that Chrysin( ChR) inhibits the capacity of self-renewal in sphere-forming cells( SFCs) derived from lung cancer A549 cell line and its potential mechanism. Methods SFCs derived from lung cancer A549 cell line were cultured in vitro. Cells were treated with various concentrations of ChR. The capacity of Self-renewal was detected by tumor sphere-forming assay. The protein expression of CK2αand Gli1 were analyzed using western blot. We used transfection with pcDNA3. 1-CK2α to explore the mechanism of those effects. Results After treated with chrysin( 10,20 and 40 μM) for 24 h,the rate of tumor sphere-forming of SFCs was significantly decreased,and the protein expression levels of CK2α and Gli1 were down-regulated in a concentration-dependent manner( P < 0. 05). Compared with the cells transfected with pcDNA3. 1-LacZ,the expression of CK2α and Gli1 were up-regulated and the rate of tumor sphere-forming was increased in SFCs that transfected with pcDNA3. 1-CK2α. And the transfection with pcDNA3. 1-CK2α can attenuated the effect of ChR on SFCs. Conclusion The effects that ChR inhibit self-renewing capacity of A549 SFCs are associated with down-regulation of CK2α and Gli1 expression.
