摘要
目的:研究紫花牡荆素(casticin,CAS)是否抑制卵巢癌OVCAR-3细胞系肿瘤球形成能力。方法:肿瘤球形成法检测不同浓度CAS(1.0、3.0、10.0μmol/L)对OVCAR-3细胞系肿瘤球形成能力的影响。Western Blot分析探讨CAS影响OVCAR-3细胞系肿瘤球形成机制是否涉及其下调干细胞标志物CD44蛋白表达。结果:CAS以浓度依赖方式抑制OVCAR-3细胞系肿瘤球形成能力。与亲本细胞相比,OVCAR-3细胞系球形成细胞高表达CD44蛋白。CAS显著下调OVCAR-3细胞系球形成细胞CD44蛋白表达。结论:CAS抑制卵巢癌OVCAR-3细胞系肿瘤球形成与其下调干细胞标志物CD44蛋白表达相关。
Objective To investigate whether casticin inhibits capability of tumorsphere formation in human ovarian cancer OVCAR-3 cells. Methods Tumorsphere formation assay was used to determine the effects of various concentrations of casticin( 1. 0,3. 0,10. 0 μmol /L) on capability of tumorsphere formation of OVCAR-3 cell line.Western blot analysis was used to probe the mechanism by whether casticin affects tumorsphere formation of OVCAR-3 cells is associated with downregulation of stem cell marker CD44 protein. Results Casticin significantly inhibited capability of tumorsphere formation of human ovarian cancer OVCAR-3 cells,in a concentration-dependent manner.Over-expression of CD44 protein existed in sphere-forming cells of human ovarian cancer OVCAR-3 cells,compared with the parental cells. Casticin could efficiently reduce the expression levels of CD44 protein in sphere-forming cells of ovarian cancer OVCAR-3 cell line. Conclusion The inhibitory effect of casticin on tumorsphere formation of human ovarian cancer OVCAR-3 cells is associated with downregulation of stem cell marker CD44 protein.
