摘要
为了研究pH值变化对猪精液液态保存效果的影响,找到适合猪精液液态保存pH值,从而改进液态保存液的配方,提高人工授精效率,试验比较了不同pH值条件下猪精液液态保存效果,通过检测精子活力、存活时间、顶体完整性和质膜完整性等参数,研究pH值变化对猪精液液态保存效果的影响,并尝试改进保存缓冲液。结果表明:采用Androhep保存时,最佳初始保存pH值为6.40。在保存的第3天稳定pH值组能够显著提高精子活力(P<0.05),用2-吗啉乙磺酸(MES)代替Androhep保存液中的4-羟乙基哌嗪乙磺酸(HEPES),在保存的第3天能显著提高精子活力、存活时间(P<0.05),同时维持更高的质膜完整性,但是顶体完整性差异不显著(P>0.05)。说明采用能够稳定pH值的保存液更有利于精液常温保存。
To study the effect of pH value changes on liquid storage of boar semen,and to find out a suitable pH value for liquid boar semenpreservationwhich could improve the ingredients of liquid storage and could enhance artificial insemination efficiency,the effect of different pH value conditions on liquid storage of boar semen was compared in this experiment. By means ofmonitoring the factors including sperm motility,sperm survival time,acrosome and membrane integrityand other factors,the effect of pH value changes on liquid storage of boar semen was studied and the improvement of liquid storage was attempted. The results showed that the best initial preservation pH was 6.40 when the Androhep was used as storage liquid; andstable pH value could significantly improvesperm motility( P<0. 05) on day 3 post semen storage. When 2-morpholinoethanesulfonic acidinstead of 4-hydroxyethylpiperazine ethane sulfonic acid( HEPES) which was contained in the Androhep storage liquid,sperm motilityand survival time were improved significantly( P < 0. 05),and meanwhile membrane integrity was maintained betteron day 3 post semen storage,but there was no significant difference in acrosomeintegrity( P>0. 05). The results verified that storage liquidwith stable pH value could be more suitable for semen preservation at room temperature.
