电针及天麻素降低脑缺血大鼠杏仁中央核Nago-A及其受体的免疫组织化学表达 被引量:2

摘要 目的观察电针及天麻素对脑缺血大鼠杏仁中央核(central nucleus of amygdale,CeA)神经轴突生长抑制因子A(neurite outgrowth inhibitor-A,Nogo-A)及其受体(Nogo-A receptor,NgR)水平的影响,了解Nago-A及其受体是否参与针药结合治疗脑缺血作用。方法将SD大鼠随机分为正常组、模型组、电针组、天麻素组和电针+天麻素组。除正常组,其余组采用线栓法复制局灶性脑缺血大鼠模型。待造模成功大鼠清醒后,模型组不予治疗,电针组电针(频率2Hz)刺激左侧"曲池""合谷"穴,天麻素组腹腔注射天麻素注射液,电针+天麻素组给予电针和天麻素联合治疗;均为每天1次,共治疗14d。采用免疫组织化学方法检测CeA内Nogo-A、NgR蛋白的水平。结果模型组Nogo-A、NgR免疫组织化学表达较正常组明显增强,电针组、天麻素组和电针+天麻素组Nogo-A和NgR表达性较模型组明显减弱,电针+天麻素组Nogo-A、NgR表达较电针组或天麻素组减弱。结论电针及天麻素均可抑制脑缺血后CeA内Nogo-A、NgR水平的升高,二者结合具有协同作用。 Objective To observe the effect of electroacupuncture(EA) and gastrodin on the level of Nogo-A and Nogo-A receptor(NgR) in the central nucleus of amygdala(CeA) of rats with cerebral ischemia(CI),so as to explore whether Nago-A and its receptors are involved in the treatment of cerebral ischemia by EA and gastrodin.Methods SD rats were randomly divided into 5groups:normal,model,EA,gastrodin,and EA + gastrodin.The CI model was established by thread occlusion in each group except the normal group.After the model rats awaked,the model group was not treated,and the others were treated once a day for a total of14 days.The EA group was stimulated on the left side 'Quchi'(LI 11) and 'Hegu'(LI 4),the gastrodin group received gastrodin injection,while the EA + gastrodin group was given EA combined with gastrodin injection.The levels of Nogo-A and NgR in CeA were detected by immunohistochemical methods.Results Compared with those of the normal group,the immunohistochemical expression of Nogo-A and NgR in the model group were significantly increased.The expression of Nogo-A and NgR were down-regulated in the EA,gastrodin and EA + gastrodin groups in comparison with those of the model group,and were decreased more significantly in the EA +gastrodin group than in the EA or gastrodin group.Conclusion EA and gastrodin down-regulates the levels of Nogo-A and NgR in CeA of CI rats,while the combined use has a synergistic effect.
