摘要
目的 探索高致瘤性K5 6 2 -n细胞系在胸腺缺陷裸小鼠体内致瘤的细胞生物学机理。方法 通过极限稀释法建立K5 6 2 -n细胞系致瘤率不同的克隆株 ,并以K5 6 2细胞为对照 ,对各克隆株进行裸小鼠体内外生物学特性的比较研究。结果 高致瘤性的K5 6 2 -B、K5 6 2 -C、K5 6 2 -E克隆株 (致瘤率≥ 5 / 6 ) ,其软琼脂培养集落形成率 ,特别是大集落形成率 ,较对照K5 6 2细胞显著增高 ,(P <0 .0 1) ,而无致瘤性或低致瘤性的K5 6 2 -A、K5 6 2 -D克隆株 (致瘤率≤ 2 / 6 ) ,其集落形成率与对照无显著差别 (P >0 .0 5 ) ;超微结构观察提示高致瘤性克隆株 ,常染色质成分增多 ,异染色质少见 ,胞浆内微丝增多且排列紊乱 ;流式细胞仪细胞周期分布分析显示高致瘤性的克隆株S期细胞比例增加。实验结果还表明裸小鼠NK细胞对高致瘤性克隆株的杀伤活性显著低于对照的K5 6 2细胞 (P <0 .0 1) ,而低致瘤性克隆株与对照无显著差别 (P >0 .0 5 ) ;组织病理观察显示低致瘤性的克隆株及对照K5 6 2细胞 ,所成裸小鼠肿瘤组织内有大量的炎性细胞浸润 ,大量肿瘤细胞被杀伤 ,血供丰富区尤甚 ,而高致瘤性克隆株所致肿瘤组织内炎性细胞浸润少见 ,肿瘤组织增殖旺盛。结论 K5 6 2 -n细胞系的裸小鼠高致瘤机理在于 :1.软琼脂集落形成率增高?
Objective To study the biological characteristics of human high tumorigenic leukemia cell line K562-n in nude mice. Me^thods Different clone strains of K562-n cells were established by limited dilution. The biological features of different clone strains were compared with K562 cells.Results The colony formation percentages in soft agar, especially the large colony formation percentages of the high tumorigenic clone strains K562-B, K562-C, K562-E were significantly higher than that in K562 cells ( P <0.01). There were no significant differences between the low or non-tumorigenic clone strains K562-A, K562-D and K562 cells ( P >0.05). Ultrastructure observation of high tumorigenic cells illustrated the euchromatic element of nuclear chromatin is increased and the heterochromatin is very rare, the microfilaments in cytoplasm are increased and the arrangement was disturbed as compared with K562 cells. The cell cycle analysis by flow cytometer showed higher S phase fractions in high tumorigentic cells. The study also showed that the killing activities of NK cells of the high clone strains are significant lower than the contrast ( P <0.01). The histopathology of nude mice tumors produced by low tumorigenic leukemia cells showed there were many inflammatory cells, mainly lymphocytes, infiltrated and many tumor cells were killed especially in vessel abundant areas. In the contrast, there were rarely inflammatory cells infiltrated in the tumors produced by high tumorigenic cell strains.Conclusion In contrast to K562 cell line, the cytobiological characteristics of high tumorigenic K562-n cell line are higher colony formation percentages in soft agar, higher S phase fraction, increased euchromatic and very rare heterochromatin, increased and disorderly arranged microfilaments in cytoplasm, decreased susceptibility to nude mice NK cells, less inflammatory cells infiltrated in the tumors produced in nude mice.
