摘要
研究旨在通过筛选皂角刺总黄酮纯化工艺,并评估纯化前后抗结肠癌的药效活性。通过测定总黄酮的吸附率和解吸率筛选合适的大孔吸附树脂型号,并采用单因素实验优化皂角刺总黄酮的纯化工艺,随后,利用CCK-8法测定其对人结肠癌细胞系SW620细胞的抑制率,比较总黄酮纯化前后的抗肿瘤活性。选取HPD-400大孔吸附树脂,皂角刺提取物以0.1 g/mL的生药浓度进行上样,流速为3 BV/h,上样体积为12 BV,5 BV水除杂,6 BV的95%乙醇洗脱,流速为3 BV/h,一次纯化与二次纯化纯度及收率分别为78.16%、86.07%,3.51%、2.89%,对SW620细胞的IC50值分别为131.23μg/mL、95.54μg/mL。优化的皂角刺总黄酮纯化工艺稳定、可靠,皂角刺总黄酮纯化物对结肠癌细胞具有抑制作用,为皂角刺的中药新药开发奠定了基础。
The purification process of total flavonoids from Saponaria sinensis was screened,and its anti-colon cancer pharmacodynamic activity before and after purification was evaluated.The adsorption rate and desorption rate of total flavonoids were used to screen the macroporous adsorption resin models,and the purification process of total flavonoids from Saponaria sinensis was optimized by single factor experiment,and the cell suppression rate of human colon cancer cell line SW620 cells was determined by CCK-8 method,and the anti-tumor activity of total flavonoids before and after purification was compared.HPD-400 macroporous adsorption resin,Saponaria sinensis extract was loaded at 0.1 g/mL crude drug concentration,flow rate of 3 BV/h,sample volume of 12 BV,5 BV water impurities,6 BV of 95%ethanol elution,flow rate of 3BV/h,the purity and yield of primary and secondary purification were 78.16%,86.07%,3.51%,2.89%,respectively,and the IC50 values of SW620 cells were 131.23μg/mL and 95.54μg/mL.The optimized purification process of total flavonoids from Saponaria sinensis is stable and reliable,and the purification of total flavonoids from Saponaria sinensis has an inhibitory effect on colon cancer cells,which lays a foundation for the development of new traditional Chinese medicine drugs for Saponaria sinensis.
