RNF175在卵巢癌中的表达及对卵巢癌细胞功能的影响

Expression of RNF175 in ovarian cancer and its impact on ovarian cancer function

摘要 目的探究环指蛋白175(RNF175)在卵巢癌中的表达及对卵巢癌细胞功能的影响。方法基于生物信息学技术分析RNF175与卵巢癌发病及预后的关系;免疫组化实验分析卵巢癌组织与癌旁组织中RNF175表达差异;细胞计数试剂盒-8及流式细胞术等分析RNF175表达对卵巢癌细胞功能的作用,基于反转录聚合酶链反应及免疫印迹法手段分析RNF175在卵巢癌细胞中的表达。结果RNF175在卵巢癌癌组织及细胞中的表达水平显著高于癌旁组织/正常细胞(P<0.05);RNF175敲低癌症组细胞增殖及迁移率显著低于癌症组,凋亡率显著高于癌症组(P<0.05)。结论相较于正常卵巢组织及细胞,RNF175在卵巢组织及细胞中存在高表达,RNF175为卵巢癌的促癌基因,可作为卵巢癌的生物标志物。 Objective To explore the expression of ring finger protein 175(RNF175)in ovarian cancer and its impact on ovarian cancer cell functions.Methods Bioinformatics analysis was conducted to analyze the relationship between RNF175 and the onset and prognosis of ovarian cancer.Immunohistochemistry experiments were used to analyze the difference in RNF175 expression between ovarian cancer tissues and adjacent normal tissues.Cell counting kit-8 and flow cytometry were used to analyze the effect of RNF175 expression on ovarian cancer cell function.Reverse transcription polymerase chain reaction and Western blotting were used to analyze the expression of RNF175 in ovarian cancer cells.Results The expression level of RNF175 was significantly higher in ovarian cancer tissues and cells compared to adjacent tissues/normal cells(P<0.05).Cell proliferation and migration rate of RNF175 knockdown cancer group were significantly lower than that of cancer group,and apoptosis rate was significantly higher than that of cancer group.Conclusion Compared to normal ovarian tissues and cells,RNF175 is highly expressed in ovarian tissues and cells.RNF175 is an oncogene in ovarian cancer and can serve as a biomarker for ovarian cancer.
