摘要
环指蛋白(RNF)180基因是一种抑癌基因,其表达水平受基因甲基化修饰的影响。RNF180基因编码产物具有E3泛素蛋白连接酶活性,参与机体细胞的生长、成熟、增殖及凋亡。目前,全球胃癌发病率及病死率较高,且尚缺乏高敏感性、高特异性的简便检测手段。外周血RNF180基因甲基化检测在胃癌早期诊断中的敏感性及特异性均较高,与肿瘤大小、TNM分期、分化程度相关,但目前对RNF180的认识尚不全面,进一步探索RNF180的作用机制、优化基因甲基化检测手段,可为胃癌早期筛查、诊断、靶向治疗、疗效评价、预后和复发监测提供新线索。
Ring finger protein(RNF)180 is a tumor suppressor gene,and its expression level is regulated by DNA methylation.The encoded protein of RNF180 is a kind of E3 ubiquitin ligase and is involved in cell development,maturation,proliferation and apoptosis.The morbidity and mortality of gastric cancer are high in the world,and there is a lack of simple detection methods with high sensitivity and specificity.Methylation detection of RNF180 gene in peripheral blood has high sensitivity and specificity for early gastric cancer detection,and is associated with tumor size,TNM stage,and degree of differentiation,although knowledge of RNF180 is still limited.Further exploration of the mechanism of RNF180 and optimization of gene methylation detection methods can provide new clues for the early screening,diagnosis,targeted therapy,efficacy evaluation,prognosis and recurrence monitoring of gastric cancer.
