摘要
目的考察山奈酚对小鼠干眼模型的体内药效学研究。方法用药物结合干燥环境诱导建立小鼠干眼模型,随机将小鼠分为5组,分别为空白对照组(N)、模型组(M)、阳性药组(Y)、山奈酚低浓度组(S1)、山奈酚高浓度组(S2),每组各5只。造模给药周期14天,第14天对各组小鼠进行泪液分泌量检测(SIT)、泪膜破裂时间检查(BUT)、角膜上皮损伤评分,并对其眼球及眼睑附属器进行组织病理学检查,考察角膜上皮层厚度、结膜上皮杯状细胞密度以及角结膜上皮细胞凋亡情况。结果与模型组相比,山奈酚低浓度组在SIT、角膜上皮损伤评分、结膜上皮杯状细胞密度以及结膜上皮细胞凋亡,具有显著性差异(P<0.05)。山奈酚高浓度组在SIT、角膜上皮损伤评分、角膜上皮层厚度、结膜上皮杯状细胞密度以及角结膜上皮细胞凋亡,均具有显著性差异(P<0.05)。结论山奈酚能够减轻由干眼引起的小鼠眼表损伤,增加泪液分泌量,并能够抑制角膜上皮细胞和结膜上皮细胞凋亡,改善角膜屏障功能。
OBJECTIVE To investigate the pharmacodynamics of kaempferol in dry eye disease(DED)model in vivo.METHODS We established a mouse DED model induced by drugs and dry environment.The mice were randomly divided into 5 groups:normal control group(N),model group(M),positive drug group(Y),low dose of kaempferol group(S1),and high dose of kaempferol group(S2),with 5 mice in each group.On the 14th day of the 14 day model cycle,schirmer I test(SIT),tear breakup time(BUT),corneal fluorescein staining scores were performed on each group of mice.Histopathological examinations were also performed on eyeballs and eyelid appendages to investigate the thickness of the corneal layer,density of conjunctival goblet cells,and apoptosis of corneal and conjunctival epithelial cells.RESULTS Compared with the model group,SIT and density of conjunctival goblet cells were significantly increased,Corneal fluorescein staining scores and number of conjunctival epithelial cell apoptosis were significantly reduced in S1 group(P<0.05).The result in S2 group showed significant differences in SIT,corneal fluorescein staining scores,corneal layer thickness,density of conjunctival goblet cells,and apoptosis of corneal and conjunctival epithelial cells(P<0.05).CONCLUSION Kaempferol can alleviate eye surface damage caused by DED,increase tear secretion,inhibit apoptosis of corneal and conjunctival epithelial cells,and improve corneal barrier function.
