兔蒸发过强型干眼白细胞介素6、肿瘤坏死因子α和黏附分子1的表达 被引量:6

Experimental study of the expression of interleukin-6,tumor necrosis factor-α and intercellular adhesion molecular-1 on evaporative dry eye in the rabbit

摘要 目的探讨炎性细胞因子在蒸发过强型干眼的发病机制中所起的作用。方法20只新西兰大白兔被随机分为实验组和对照组两组,实验组烧灼兔睑板腺开口制成蒸发过强型干眼模型,对照组不作处理。第1、第6、第11周用放射免疫法及酶联免疫法测泪液中白细胞介素6(interleukin-6,IL-6)和肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)的含量,第11周时处死兔,用免疫组化方法检测细胞间黏附因子1(intercellular adhesion molecule-1,ICAM-1)在角膜、结膜组织中的表达。结果实验组兔SchirmerⅠ滤纸湿长、泪膜破裂时间均较正常组增加(P<0.01);实验组泪液中IL-6、TNF-α的含量均较对照组增高(P<0.01);ICAM-1在两组兔角膜、结膜组织上皮均有表达,阳性反应物呈棕黄色,主要表达于细胞浆。在实验组表达较强,在对照组表达较弱,两者差异有非常显著的统计学意义(P<0.01)。每1000个细胞中阳性细胞表达数在实验组(角膜为272.1±46.8,结膜为302.7±56.3)明显高于对照组(角膜为50.2±5.9,结膜为60.4±15.3)(P<0.01)。结论细胞因子在蒸发过强型干眼的发病机制中起着重要的作用,蒸发过强型干眼的发病机制与炎症有关。 Objective To investigate the role of inflammatory cytokine in evaporative dry eye. Methods Twenty rabbits were randomly divided into an experimental group and a control group. A dry eye model was established by cauterizing the meibomian gland orifices of rabbits in the experimental group. ELISA and radioimmunity were used to detect the concentrations of interleukin-6 (IL- 6) and tumor necrosis factor-alpha (TNF-α in tears at 1, 6, and 11 weeks after cauterizing the meibomian gland. The rabbits were sacrificed 11 weeks after cauterizing the meibomian gland. Their corneal and conjunctival epithelia were examined by immunohistochemical staining to detect the expression of intercellular adhesion molecular-1 (ICAM-1). Results Schirmer Ⅰ test wetting in the experimental group (5.1±1.6)mm was significantly shorter than that of the control group (18.6±4.2)mm at 6 weeks after cauterizing (P〈 0.01). BUT time in the experimental group (3.4±1.1)s was shorter than that of the control group (17.6±3.4)s (P〈0.01). The concentration of IL-6 in tears was higher in the experimental group (434.4±38.7)pg/ml than in the control group (150.1±23.6)pg/ml (P〈0.01). The concentration of TNF-α in tears was higher in the experimental group (7.63±0.40)pg/m] than in the control group (3.53±0.54)pg/ml (P〈0.01). ICAM-1 expression in positive cells of 1000 cells in the corneal and conjunctival epithelia in the experimental group (272.1±46.8, 302.7±56.3, respectively) was stronger than that of the control group (50.2±5.9, 60.4±15.3, respectively) (P〈0.01). Conclusion Cytokines play an important role in the pathogenesis of evaporative dry eye.
