基于重测序的‘蒙冠1号’、‘蒙冠2号’全基因组遗传变异分析

Genetic Variation Analysis of the Whole Genome of'Mengguanl'and'Mengguan2'Based on Resequencing

摘要 为了研究不同文冠果品种之间的遗传变异,采用高通量重测序技术,对两个高产文冠果品种‘蒙冠1号’‘蒙冠2号’分别进行了全基因组测序,平均测序深度为93.35×和86.76×,覆盖率为99.96%,并与参考基因组比对,在两个品种中检测到了丰富的遗传变异,包括SNP和InDel。对存在遗传变异的基因进行GO富集,发现它们主要富集在细胞壁合成和胁迫响应。针对不同品种的差异,发现‘蒙冠1号’主要富集在细胞壁生物发生及生长发育通路,‘蒙冠2号’主要与胁迫响应和防御通路相关,推测这些基因的遗传变异是不同性状差异的原因。本研究结果一定程度上反映了文冠果两个品种之间在基因组水平的差异,为分子标记的开发提供了遗传基础,也为后续的分子育种及品种改良提供了理论指导和科学依据。 In order to study the ge netic variation among different variations of Xanthoceras sorbifolium Bunge species, the whole genomes of two high-yielding 'Mengguan1' and 'Mengguan2' were sequenced using high throughput resequencing technology. The average sequencing depth was 93.35× and 86.76×, and the coverage rate was 99.96%. Compared with reference genome, rich genetic variations, including SNP and InDel, were detected in both cultivars. GO enrichment was performed on genes with genetic variation, and it was found that they were mainly enriched in cell wall synthesis and stress response. According to the differences among different varieties, 'Mengguan1' was mainly enriched in the cell wall biogenesis, growth and development pathways, while 'Mengguan2' was mainly related to stress response and defense pathways. It was speculated that the genetic variation of these genes was the reason for different traits. To some extent, the results of this study reflect the differences between the two varieties at the genomic level, which provides the genetic basis for the development of molecular markers, and also provides theoretical guidance and scientific basis for the subsequent molecular breeding and variety improvement.
