诃子清幽方提取工艺优化及体外抗幽门螺杆菌活性研究

Optimization of the Extraction Process and in vitro Anti-Helicobacter pylori Activity of Hezi Qingyou Formula

摘要 在证明诃子清幽方的主要成分没食子酸和鞣花酸具有抗幽门螺杆菌活性的基础上,以加水量、提取时间、提取次数为影响因素,没食子酸和鞣花酸含量以及提取率为评价指标,采用正交试验优选诃子清幽方的提取工艺,并考察水提物对幽门螺杆菌的体外抑制活性。结果显示,诃子清幽方的优化提取工艺为:加水量10倍,提取时间1.5 h,提取次数2次。在该条件下,诃子清幽方水提物在体外对幽门螺杆菌生长有显著的抑制作用且呈现浓度依赖性,MIC为80~160μg/mL。 In this study,based on the observation of the activity of both gallic acid and ellagic acid against Helicobacter pylori(H.pylori)in vitro,an orthogonal test was used to optimize the extraction process of Hezi Qingyou formula,with the adding amount of water,extraction time and extraction times as influencing factors,content of gallic acid and ellagic acid and extraction rate as evaluation indexes.And the in vitro inhibitory activity of the aqueous extracts against H.pylori was investigated.The results showed that the optimized extraction process of Hezi Qingyou formula was as follows:adding 10 times amount of water,extraction 1.5 h per time and 2 times of extraction.Under these conditions,the aqueous extracts of Hezi Qingyou formula showed significant in vitro inhibition of H.pylori growth in a concentration-dependent manner,with the MIC of 80-160μg/mL.
