摘要
目的:为探讨丹阳地区结直肠癌(colorectal cancer,CRC)患者外周血中LC3B^(+)EVs与髓源性抑制细胞(myeloid-derived suppressor cell,MDSC)及各细胞因子的水平及其临床相关性。方法:流式细胞仪分别检测两组健康体检者和CRC患者外周血中LC3B^(+)EVs、MDSC及粒系髓源性抑制细胞(PMN-MDSC)和单核系髓源性抑制细胞(M-MDSC)的水平,讨论LC3B^(+)EVs与MDSC的临床相关性,并将CRC患者的血浆标本中的效应分子进行多因子检测(细胞因子芯片),讨论LC3B^(+)EVs与各因子的相关性。结果:与对照组相比,CRC患者外周血LC3B^(+)EVs数量显著上升(P<0.0001),且与患者年龄、性别无统计学差异(P=0.8690,P=0.9712)。CRC患者外周血中MDSC及PMN-MDSC、M-MDSC两个亚群的水平升高皆有统计学意义,并以PMN-MDSC增高为主(P<0.0001)。CRC患者外周血中LC3B^(+)EVs与MDSC及PMN-MDSC的水平无相关性(r=-0.002699,P=0.9891;r=-0.04435,P=0.8227),但与M-MDSC的水平呈显著正相关(r=0.4811,P=0.0096)。CRC患者外周血LC3B^(+)EVs与IL-1β、IL-2、IL-4、IL-8、IL-17、IFN-γ的水平呈正相关(r=0.7031、r=0.7827、r=0.4663、r=0.5901、r=0.4589、r=0.4655,P<0.05),尤其是与IL-1β呈显著正相关(P=0.0016);而与IL-5、IL-6、IL-10、IL-12、TNF-α的水平无统计学意义(P>0.05)。结论:CRC患者血浆中LC3B^(+)EVs数量与MDSC水平显著高于健康人,LC3B^(+)EVs与M-MDSC亚群及IL-1β的水平存在显著正相关,提示M-MDSC与IL-1β可能诱导CRC肿瘤细胞释放自噬小体增加,它们之间可能存在相互作用从而影响CRC的发生发展。
Objective:To investigate the levels and clinical correlations of LC3B^(+)EVs,myeloid-derived suppressor cell(MDSC),and various cytokines in peripheral blood of colorectal cancer(CRC)patients in Danyang area.Methods:Flow cytometry was used to detect LC3B^(+)EVs and the levels of MDSC,PMN-MDSC and M-MDSC in peripheral blood of two groups of healthy individuals and CRC patients,and to discuss the clinical correlation between LC3B^(+)EVs and MDSC.And multi factor detection was performed on the effector molecules in the plasma samples of CRC patients,and the correlation between LC3B^(+)EVs and various factors was discussed.Results:Compared with the control group,the number of LC3B^(+)EVs in the peripheral blood of CRC patients significantly increased(P<0.0001),and there was no statistically significant difference with the patient's age and gender(P=0.8690,P=0.9712).There was a statistically significant increase in MDSC,PMN-MDSC,and M-MDSC in the peripheral blood of CRC patients,with PMN-MDSC mainly increasing(P<0.0001).There was no correlation between the levels of LC3B^(+)EVs and MDSC and PMN-MDSC in peripheral blood of CRC patients(r=-0.002699,P=0.9891,r=-0.04435,P=0.8227),but were significantly positively correlated with M-MDSC(r=0.4811,P=0.0096).LC3B^(+)EVs in peripheral blood of CRC patients with IL-1β,IL-2,IL-4,IL-8,IL-17 and IFN-γwere positively correlated(r=0.7031,r=0.7827,r=0.4663,r=0.5901,r=0.4589,r=0.4655,P<0.05),especially with IL-1β,there was a significant positive correlation(P=0.0016).And there was not a significant positive correlation with the level of IL-5,IL-6,IL-10,IL-12,TNF-α(P>0.05).Conclusion:The number of LC3B^(+)EVs and MDSC levels in the plasma of CRC patients are significantly higher than those of healthy individuals.There is a significantly positive correlation between LC3B^(+)EVs and the accumulation of M-MDSC subgroups and IL-1β,which may induce increased release of autophagosomes from CRC tumor cells,and there may be interactions between them that affect the occurrence and developme
