电针干预对单眼形觉剥夺弱视大鼠视皮层突触结构及视觉诱发电位的影响

Effects of Electro-Acupuncture on Synaptic Structure and Visual Electrophysiological Function of Visual Cortex in Monocular Form Deprivation Amblyopia Rats

摘要 目的探讨电针刺激对单眼形觉剥夺Long Evans大鼠视皮层中蛋白激酶C zeta(protein kinase C zeta,PKCζ)、突触素(synaptophysin,Syn)及突触后致密蛋白95(postsynaptic density-95,PSD-95)表达影响,观察电针干预对弱视眼视觉电生理功能改变及初级视皮层中星形胶质细胞数量的改变。方法选取48只Long Evans大鼠新生幼鼠,随机分为正常对照组、弱视模型组、电针治疗组、假穴治疗组。正常对照组不做任何处理,弱视模型组、电针治疗组及假穴治疗组对幼鼠右眼单眼缝合建立弱视模型,电针治疗组和假穴治疗组在生后30 d开始电针刺激。采用闪光视觉诱发电位(flash visual evoked potential,F-VEP)对各组大鼠右眼视觉电生理功能进行检测;应用蛋白免疫印迹技术(western blotting,WB)检测大鼠左侧初级视皮层V1M中PKCζ、Syn及PSD-95的表达量;借助免疫荧光技术和星形胶质细胞特异性标志物胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)观察大鼠左侧初级视皮层V1M区中星形胶质细胞数量的变化。结果F-VEP结果显示,与正常对照组相比,弱视模型组或假穴治疗组P2波幅明显降低(P<0.01),P2潜伏期可见延长(P<0.05),电针治疗组P2波幅略低(P<0.05),P2潜伏期差异无统计学意义;与弱视模型组或假穴治疗组相比,电针治疗组的P2波幅增加(P<0.05),P2潜伏期变短(P<0.05)。蛋白免疫印迹结果显示,与正常对照组相比,弱视模型组或假穴治疗组PKCζ表达量减少(P<0.05),Syn及PSD-95表达均明显降低(P<0.01),电针治疗组PKCζ、Syn及PSD-95表达差异无统计学意义;与弱视模型组或假穴治疗组相比,电针治疗组PKCζ、Syn及PSD-95表达均明显增加(P<0.05);免疫荧光结果显示,与正常对照组相比,弱视模型组或假穴治疗组中GFAP阳性面积显著减少(P<0.01),而电针治疗组中GFAP阳性面积差异无统计学意义;与弱视模型组或假穴治疗组相比,电针治疗组中GFAP阳性面积明显增� Objective To investigate the effect of electro-acupuncture on the expressions of protein kinase C zeta(PKCζ),synaptophysin(Syn)and postsynaptic density-95(PSD-95)in the visual cortex of monocular form deprived Long Evans rats and observe the changes of visual electrophysiological function of amblyopia and the number of astrocytes in the visual cortex.Methods Forty-eighty newborn Long Evans rats were randomly divided into normal control group,amblyopia model group,electro-acupuncture treatment group and sham acupoint treatment group.The normal control group did not do any treatment.The amblyopia model group,electro-acupuncture treatment group and sham acupoint treatment group sutured the right eye of young rats to establish the amblyopia model.The electro-acupuncture treatment group and sham point treatment group began electro-acupuncture stimulation 30 days after birth.Flash visual evoked potential(F-VEP)was used to detect the visual electrophysiological function of the right eye of rats in each group,and Western blotting(WB)was used to detect the expressions of PKCζ,Syn and PSD-95 in V1M of left visual cortex of rats.The number of astrocytes in the V1M area of the left visual cortex of rats was observed by immunofluorescence and glial fibrillary acid protein(GFAP).Results The results of F-VEP showed that compared with that of the normal control group,the amplitude of P2 in the amblyopia model group or sham acupoint treatment group was significantly lower(P<0.01),the latency of P2 was significantly prolonged(P<0.05),and the amplitude of P2 in the electro-acupuncture treatment group was slightly lower(P<0.05)and there was no statistical difference in the latency of P2.Compared with those of the amblyopia model group or sham acupoint treatment group,the P2 amplitude of the electro-acupuncture treatment group increased(P<0.05),and the P2 latency became shorter(P<0.05).WB showed that compared with those of the normal control group,the expression of PKCζin amblyopia model group or sham acupoint treatment group decr
