2020-2021年我国规模化猪场猪圆环病毒2型和3型的分子流行病学调查分析 被引量:3

Molecular Epidemiological Investigation and Analysis of Porcine Circovirus Type 2 and Porcine Circovirus Type 3 in Large-scale Pig farms in China from 2020 to 2021

摘要 采集13个省市自治区的491个规模化猪场的1447份疑似猪圆环病毒感染样本,通过荧光定量PCR方法进行猪圆环病毒2型(PCV2)和猪圆环病毒3型(PCV3)的检测。分别随机选取50份PCV2和PCV3阳性样品,对其ORF2基因进行测序并分别进行核苷酸同源性比对和遗传进化分析。结果显示,样本中PCV2和PCV3检出率分别为29.16%(422/1447)和18.93%(274/1447)。规模化猪场中PCV2和PCV3检出率分别为47.05%(231/491)和15.68%(77/491),PCV2和PCV3混合感染率为23.81%,混合感染高于单一感染,与猪繁殖与呼吸综合征病毒(PRRSV)、伪狂犬病病毒(PRV)、猪肺炎支原体(MhP)等病原体以多种组合的混合感染形式出现。50个PCV2序列与16个参考毒株序列的核苷酸和氨基酸序列同源性分别为75.6%~100%和83.1%~99.1%,PCV3对应同源性分别为95.9%~100%和96.3%~100%。分子遗传进化分析显示PCV2d占80%(40/50),PCV2a占4%(2/50),PCV2b占16%(8/50),未检测到PCV2c和PCV2e。PCV3a、PCV3b和PCV3c分别占28%(14/50)、22%(11/50)和50%(25/50)。表明2020-2021年我国规模化猪场PCV2的主要流行基因型是PCV2d,其次为PCV2a和PCV2b。PCV3c为我国规模化猪场中的主要流行基因型,而PCV3a和PCV3b流行率也依然较高。论文对我国猪圆环病毒的分子流行病学进行了数据补充,对临床防控起到了预警作用,也为猪圆环病毒疫苗的研发提供参考。 In this study,1447 samples of suspected porcine circovirus infection were collected from 491 large-scale pig farms in 13 provinces and autonomous regions,and PCV2 and PCV3 were detected by fluorescence quantitative PCR method.50 from each of PCV2 and PCV3 positive samples were randomly selected and their ORF2 genes were sequenced,respectively.Nucleotide homology alignment and genetic evolutionary analysis were performed using MegAlign and MEGA6 software,respectively.The results showed that the detection rates of PCV2 and PCV3 in the samples were 29.16%(422/1447)and 18.93%(274/1447),respectively.Detection rates of PCV2 and PCV3 in large-scale pig farms were 47.05%(231/491)and 15.68%(77/491),respectively.The mixed infection rate of PCV2 and PCV3 was 23.81%,which was much higher than their respective single infection rates and appeared in the form of mixed infection with pathogens such as porcine reproductive and respiratory syndrome virus(PRRSV),pseudorabies virus(PRV)and Mycoplasma hyopneumoniae(MhP)in various combinations.Nucleotide and amino acid sequence homologies of the 50 PCV2 sequences to the 16 reference strains ranged from 75.6%to 100%and 83.1%to 99.1%,respectively,and the corresponding homologies of PCV3 ranged from 95.9%to 100%and 96.3%to 100%,respectively.Molecular genetic evolutionary analysis showed that PCV2d accounted for 80%(40/50),PCV2a for 4%(2/50),PCV2b for 16%(8/50),and PCV2c and PCV2e were not detected.PCV3a,PCV3b,and PCV3c accounted for 28%(14/50),22%(11/50),and 50%(25/50),respectively.The results indicated that the primary prevalent genotype of PCV2 in large-scale pig farms in China from 2020 to 2021 is PCV2d,followed by PCV2a and PCV2b.PCV3c is the primary predominant genotype in large-scale pig farms in China,while the prevalence of PCV3a and PCV3b is still high.This study complements the data on the molecular epidemiology of porcine circovirus in China.It provides an early warning for its clinical prevention and control,as well as a reference basis for developing a porcine circovirus vac
