摘要
为了解2021年我国猪圆环病毒2型(PCV2)流行情况,本试验于7个地区16个省采集1685份组织样品,采用实时荧光定量PCR检测方法检测PCV2,对其中45份阳性样品进行全基因组序列扩增和遗传演化分析。结果显示,被检样品PCV2总阳性率为9.85%(166/1685),场总阳性率为32.71%(35/107);各地区样品阳性率介于4.29%~19.00%,东北地区样品阳性率最高;各地区场阳性率介于6.90%~60.00%,华中地区场阳性率最高。PCV2a、PCV2b和PCV2d亚型分离毒株占比分别为31.11%、4.44%和64.44%,其中PCV2d亚型为优势流行株,未发现PCV2e和PCV2c亚型。有24株PCV2分离毒株的Cap蛋白在C末端有1个赖氨酸(K)延伸,另有1株PCV2分离毒株的Cap蛋白在C末端有1个蛋氨酸(M)延伸。结果表明,2021年我国PCV2感染范围广,污染程度高,且正在不断发生遗传变异,需要持续跟踪监测PCV2的流行情况,以便及时制定有效的防控措施。
In order to understand the prevalence of porcine circovirus type 2(PCV2)in China in 2021,a total of 1685 tissue samples were collected from 16 provinces in 7 regions in this study.Fluorescent quantitative PCR was used to detect PCV2,and 45 positive samples were selected for whole-genome sequencing and genetic evolutionary analysis.The results showed that the overall positive rate of PCV2 in the tested samples was 9.85%(166/1685),while the overall positive rate at the farms was 32.71%(35/107).The positive rate of the samples in different regions ranged from 4.29%to 19.00%,with the highest positive rate in northeast China.The positive rate of different farms in various regions ranged from 6.90%to 60.00%,with the highest positive rate in central China.The proportions of PCV2a,PCV2b and PCV2d subtypes were 31.11%,4.44%and 64.44%,respectively,with PCV2d subtype being the dominant prevalent strain.No PCV2e and PCV2c subtypes were detected.Among the isolated PCV2 strains,24 isolates had a lysine(K)extension at the C-terminal of Cap protein,and one isolate had a methionine(M)extension.This study indicates that PCV2 have a wide range of infections with high contamination rate in China in 2021,and ongoing genetic variations were observed.Continuous monitoring of PCV2 prevalence is necessary to develop timely and effective control measures.
