摘要
目的观察小檗碱(BBR)预处理对大鼠减压性肺损伤的预防作用, 探索应急减压暴露的快速干预措施。方法按照随机数字表法将50只健康雄性SD大鼠分为5组, 对照组、模型组和低、中、高剂量组。3个剂量组大鼠分别于加压前3 h腹腔注射1 ml BBR溶液(12.5、25.0、50.0 mg/kg), 对照组和模型组腹腔注射1 ml生理盐水。对照组大鼠不作处理, 模型组和剂量组大鼠在3 min内空气加压到60 m, 高压下共停留60 min, 随后(30±5)s内匀速减至常压出舱。观察大鼠出舱后30 min内的死亡率, 采用酶联免疫吸附试验(ELIASA)观察大鼠肺组织的超氧化物歧化酶(SOD)水平, 采用硫代巴比妥酸(TBA)法观察大鼠肺组织的丙二醛(MDA)水平, 采用免疫组化和TUNEL试剂盒观察大鼠肺组织的细胞凋亡情况。结果模型组和低、中、高剂量组大鼠的死亡率分别为50%、30%、30%和40%。与对照组相比, 模型组大鼠肺组织的SOD水平明显降低、MDA水平明显升高, 中剂量组的MDA水平明显升高, 差异有统计学意义(P<0.05);与模型组相比, 低、高剂量组大鼠肺组织的SOD水平明显升高, MDA水平明显降低, 差异有统计学意义(P<0.05);与低剂量组相比, 中剂量组大鼠肺组织的SOD水平明显降低, MDA明显水平升高, 差异有统计学意义(P<0.05)。与对照组相比, 模型组大鼠肺组织间质充血水肿明显;剂量组大鼠肺组织间质充血水肿程度明显比模型组轻, 剂量越高, 程度越轻, 高剂量组与对照组比较差异无统计学意义(P>0.05)。模型组大鼠肺组织的细胞凋亡率比对照组明显升高, 低剂量组比模型组明显降低, 差异有统计学意义(P<0.05)。结论 BBR预处理可快速抑制大鼠减压性肺损伤的氧化应激, 降低细胞凋亡率, 起到保护作用, 最佳预防用药剂量有待进一步探索;氧化应激程度或可作为减压损伤程度的早期预警指标。
Objective To observe the preventive effects of pretreatment with Berberine(BBR)on pulmonary decompression injury,so as to explore the rapid intervention measures for emergency decompression exposure.Methods Using the random number table method,a total of 50 healthy male SD rats were divided into five groups:control group,model group,and low,medium,and high dose BBR pretreatment groups.The rats of the three BBR pretreatment groups were intraperitoneally injected with 1 ml BBR solution with different concentrations of 12.5,25.0,and 50.0 mg/kg,respectively,three hours before pressurization.The control group and the model group were intraperitoneally injected with 1 ml normal saline.The control group rats were not treated with pressurization,while the rats in other four groups were treated with pressurization to 60 m within three minutes and continued for 60 minutes,and then rapidly decompressed to the normal atmospheric pressure within(30±5)seconds at an even speed before exiting the chamber.The mortality rate of rats within 30 minutes after exiting the chamber was observed.The level of superoxide dismutase(SOD)in the homogenate of lung tissue was observed by enzyme-linked immunosorbent assay(ELISA),the level of malondialdehyde(MDA)was measured by the thiobarbituric acid(TBA)method,and the apoptosis was measured by immunohistochemistry and TUNEL assay.Results The mortalities of the model group and the low,medium,and high dose BBR pretreatment groups were 50%,30%,30%,and 40%,respectively.Compared with the control group,the SOD level in the lung tissue of the model group rats was decreased significantly,and the MDA level was increased significantly(P<0.05).Specifically,the MDA level in the medium dose BBR pretreatment group was significantly increased,and the difference was statistically significant(P<0.05).Compared with the model group,the SOD levels of the low and high dose BBR pretreatment groups were significantly increased,while the MDA levels in both groups were significantly decreased,all with statistically si
