聚乙烯醇-磺胺嘧啶前药的合成及抗菌性能研究

摘要 将聚乙烯醇(PVA)与一氯乙酸反应得到氯乙酰聚乙烯酯(PVA-Cl),PVA-Cl再与磺胺嘧啶钠(SD-Na)反应得到一种新型高分子缓释抗菌剂聚乙烯醇接载磺胺嘧啶(PVA-SD)M。采用红外光谱、核磁氢谱和紫外光谱对PVA-SD的结构进行了表征,证明产物具有预期的结构;采用紫外分光光度法考察了PVA-SD的释药性能,绘制了前药的释药曲线。结果表明,PVA-SD在不同pH值缓冲溶液中可以缓慢、持续地释放出抗菌成分。抑菌圈试验证实,PVA-SD对大肠杆菌和金黄色葡萄球菌都有较好的抑菌效果,光密度法测得PVA-SD对大肠杆菌和金黄色葡萄球菌的最小抑菌浓度(MIC)为62.5μg/mL。 A novel polymer sustained-release antibacterial agent,polyvinyl alcohol loaded sulfadiazine(PVASD),was obtained by reacting polyvinyl alcohol(PVA)with monochloroacetic acid to obtain chloroacetyl polyethylene ester(PVA-Cl).The structure of PVA-SD was characterized by infrared spectroscopy(IR),nuclear magnetic resonance hydrogen spectroscopy(1 HNMR),and ultraviolet spectroscopy(UV),proving that the product has the expected structure;The release performance of PVA-SD was investigated using UV spectrophotometry,and the release curve of the prodrug was plotted.The results showed that PVA-SD can slowly and continuously release antibacterial components in buffer solutions with different pH values.The antibacterial circle test confirmed that PVA-SD has good antibacterial effects on both Escherichiacoli and Staphylococcus aureus.The minimum inhibitory concentration(MIC)of PVA-SD against Escherichiacoli and Staphylococcus aureus was measured by optical density method to be 62.5μg/mL.
