基于灰色关联分析的荜茇体外抗氧化活性谱效关系研究

摘要 目的研究荜茇不同极性部位高效液相色谱(HPLC)图谱与其体外抗氧化活性的谱效关系,明确荜茇抗氧化物质基础。方法建立荜茇不同极性部位的HPLC图谱,并采用HPLC-Q-Orbitrap HRMS对共有峰进行识别分析;对INS-1胰岛β细胞进行高糖损伤,建立细胞氧化损伤模型,通过荜茇不同部位干预,并采用荧光酶标仪检测细胞内的活性氧(ROS)水平,筛选荜茇的体外抗氧化活性部位;采用灰色关联度分析荜茇各极性部位HPLC特征峰与抗氧化活性之间的谱效关系,寻找对抗氧化作用贡献较大的成分。结果分别得到石油醚部位1.70 g,乙酸乙酯部位5.18 g,正丁醇部位51.21 g,水提部位3.16 g,得到胡椒碱单晶;细胞内抗氧化活性实验结果表明正丁醇部位的抗氧化活性较好,HPLC图谱表明荜茇共有18个特征峰,灰色关联度结果表明荜茇图谱中峰1(荜茇宁,piperlonguminine),峰7(荜茇环碱,pipernonaline),峰5(pipercallosine),峰6(荜茇壬三烯哌啶,dehydropipernonaline),峰4(墙草碱,pellitorin),峰3(胡椒碱,piperine),峰8(假荜茇酰胺B,retrofractamide B),峰12(brachystamide B),峰11[(2E,4E,13E)-14-(benzo[d][1,3]dioxol-6-yl)-Nisobutyltetradeca-2,4,13-trienamide]和峰10(几内亚胡椒碱,guineensine)与体内抗氧化活性密切。结论本文采用方法科学严谨,能较为全面和准确地反映荜茇中具有抗氧化活性的有效成分,为荜茇的进一步开发利用提供科学依据和理论支持。 Objective To explore the spectrum-activity relationship between high performance liquid chromatography(HPLC)chromatograms of different polar fractions from Piperis longi Fructus and their in vitro antioxidant activities,to determine the antioxidant material basis of Piperis Longi Fructus.Methods Different polar fractions from Piperis longi Fructus were prepared by solvent extraction and their characteristic HPLC fingerprints were established.The common peaks were identified by HPLC-Q-Orbitrap HRMS.The INS-1 isletβcells were subjected to high glucose damage to establish a model of cellular oxidative damage,and the ROS levels in theβcells were detected by fluorescence microplate reader to investigate the antioxidant activities of different polar fractions.The spectrum-activity relationships between HPLC characteristic peaks and antioxidant activity of different polar fractions from Piperis longi Fructus were detected with gray correlation analysis,in order to screen the components that contributed to anti-oxidation.Results 1.70 g of petroleum ether fraction,5.18 g of ethyl acetate fraction,51.21 g of n-butanol fraction,3.16 g of water fraction,and piperine single crystal were obtained.The results of intracellular antioxidant activity experiments indicated that the antioxidant activity of the n-butanol fraction was the highest.The HPLC fingerprint showed 18 characteristic peaks in Piperis longi Fructus,and the results of gray correlation analysis indicated that the peaks of 1(piperlonguminine),7(pipernonaline),5(pipercallosine),6(dehydropipernonaline),4(pellitorin),3(piperine),8(retrofractamide B),12(brachystamide B),11((2E,4E,13E)-14-(benzo[d][1,3]dioxol-6-yl)-N-isobutyltetradeca-2,4,13-trienamide)and 10(guineensine)had close relationship with antioxidant activities.Conclusion The method adopted in this paper is scientific and rigorous,which could comprehensively and accurately screen the effective components with antioxidant activity in Piperis longi Fructus,and provide scientific basis and theoretical support f
