AFS-9700型原子荧光光度计测定尿中砷的研究

摘要 目的:利用AFS-9700型原子荧光光度计建立尿中砷的测定方法。方法:尿样经硫酸、硝酸、过氧化氢处理后,将硫脲、盐酸和抗坏血酸试剂直接加入10mL具塞刻度消化管内定容后,上机测定。调整仪器条件为最佳的情况下,分析研究测定中盐酸和硼氢化钾的最适浓度、样品前处理、加标回收率、精密度及检出限等,并对比该方法与WS/T 474-2015《尿中砷的测定氢化物发生原子荧光法》的结果。结果:该文建立的测定方法中,尿砷处于0~100μg/L的含量范围时,荧光强度与砷浓度间有线性关系,检出限0.07μg/L、加标回收率95%~105%、相关系数不低于0.9995,相对标准偏差低于10%。相比标准方法而言,P>0.05,两者差异无统计学意义。结论:该文建立的方法相比标准方法而言,弥补了前处理中过多取样量、使用器材复杂、酸用量多、操作繁琐等缺点,且操作简便、检出限低、线性范围宽、基体干扰少,在低、中、高及大批量尿中砷测定中适用,可行性与应用价值可观。 Objective:To establish a method for the determination of arsenic in urine by AFS-9700 atomic fluorescence photometer.Methods:Bird samples were treated with sulfuric acid,nitric acid and hydrogen peroxide,and thiourea,hydrochloric acid and ascorbate were directly added into 10mL stopper scale tube to determine the volume.The optimal concentration of hydrochloric acid and potassium borohydride,sample pretreatment,standard recovery,precision and detection limit were analyzed and studied under optimal instrument conditions,and the results of this method were compared with those of WS/T 474-2015“Determination of Arsenic in urine by hydride generation atomic fluorescence Method”.Results:When urinary arsenic content was in the range of 0-100μg/L,the fluorescence intensity had a linear relationship with arsenic concentration.The detection limit was 0.07μg/L,the recovery rate was 95%-105%,the correlation coefficient was not less than 0.9995,and the relative standard deviation was less than 10%.Compared with the standard method,P>0.05,the difference was not statistically significant.Conclusion:Compared with the standard method,the method established in this paper can make up for the shortcomings of excessive sampling amount,complicated equipment,large amount of acid and complicated operation in pre-treatment.Besides,it is simple to operate,low detection limit,wide linear range and less matrix interference.It is suitable for the determination of arsenic in low,medium,high and large quantities of urine,and its feasibility and application value are considerable.
