湿法消解-紫外分光光度法对改性透明质酸中硒含量的测定

Determination of Selenium Content in Modified Hyaluronic Acid by Wet Digestion Ultraviolet Spectrophotometry

摘要 文章旨在建立一种对改性透明质酸(Hyaluronic acid,HA)中硒含量快速检测的湿法消解-紫外分光光度法。采用湿法消解,通过对消解液组合类型和比例、溶液pH值、邻苯二胺盐酸盐浓度及络合时间等4个参数进行优化,采用优化后的试验方案对改性HA中的硒含量进行检测,同时与目前常用的电感耦合等离子体法(Inductively coupled plasma,ICP)进行比较。结果发现,该研究建立的硒元素检测方法在0.0625-20μg/mL浓度范围内的相关系数为0.9994,检出限为0.016μg/mL,定量限为0.054μg/mL,仪器相对偏差为1.08%,加标回收率为97.0%-104.0%。最终测得改性HA样品中的硒含量为(45.51±0.26)μg/mg,与ICP法在统计学上不存在显著性差异(P>0.05)。该方法对标准品和实际样品均具有良好的消解效果,且精密度、灵敏度和准确度高,不仅适用于对改性HA中硒含量的测定,也为其他改性天然聚合物中硒含量的测定提供一种可行性的方案。 A wet digestion-ultraviolet spectrophotometry method was established for rapid quantification of selenium in modified hyaluronic acid(HA).Using wet digestion four parameters were optimized including the digestion solution type and ratio,pH,o-phenylenediamine hydrochloride concentration and complexation time.The selenium in modified HA was quantified by the optimized experiment condition and compared with the inductively coupled plasma method(IcP)which was commonly used.It was found that the correlation coefficients of the selenium quantification method established in this study were 0.9994 at the concentration range of 0.0625-20μg/mL,the detection limit was 0.016μg/mL,the quantitative limit was 0.054μg/mL,the relative standard deviation was 1.08%,and the recovery was 97.0%-104.0%.The selenium quantification in the modified HA sample was(45.51±0.26)μg/mg,there was no statistically significant difference compared with IcP method(P>0.05).This method has a good digestion effect on both the standard substance and real samples,and the precision,sensitivity and accuracy are high.It is not only suitable for the determination of selenium quantification in modified HA,but also provides a feasible scheme for the determination of selenium content in other modified natural polymers.
