摘要
目的 :探讨有效阻断视网膜新生血管发生的方法。方法 :对体外培养新生儿脐静脉血管内皮细胞进行血管内皮细胞生长因子 1 65 ( VEGF165)单克隆抗体、1 3-顺式维甲酸、INF- α、INF- γ4种药物筛选实验。用倒置显微镜观察细胞生长状态 ;用 MTT法测 A560 nm值 ,计算细胞增殖抑制率。结果 :VEGF165单克隆抗体组浓度在 2 0 0 0 mg· L-1时 ,对血管内皮细胞的增殖抑制率达 91 %。1 3-顺式维甲酸对细胞最高增殖抑制率是 61 % ;IFN- γ浓度在 2 0 0 0 mg·L-1时 ,对细胞增殖抑制率为 5 6%。IFN- α浓度在 2 0 0 0 mg·L-1时 ,细胞增殖抑制率均低于 40 %。结论
Objective: To look for effective agent to block retinal neovascular angiogenesis. Methods:Four agents including anti VEGF monoclonal a ntibodies,AILIK, IFN α and IFN γ,were cultivated with human umbilical vein endothelial cells (HuVEC),respectively. The growth of HuVEC was observed by light microscopy and A 560nm were measured with MTT, and HuVEC inhibition rate by each agent were calculated. Results:The HuVEC inhibition rate was 91%,61%,56% and 40% in Anit VEGF McAb.AILIK,IFN γ and IFN α,respectively, at concentration of 2 000 mg·L -1 .Conclusion: Anti VEGF McAb have potential inhibition effect on HuVEC growth.
