摘要
目的探讨补肺填精方对大鼠薄型子宫内膜容受性的影响。方法选取未交配10周龄SD雌性大鼠,体重180^220 g。适应性喂养7 d后,连续阴道涂片HE染色10 d,筛选40只动情周期规则的SD大鼠。随机选取6只雌性大鼠作为正常对照组,其他34只雌性大鼠建立肺肾阴虚模型,造模成功后,随机选取6只作为肺肾阴虚组,其他造模成功的大鼠建立薄型子宫内膜大鼠模型,造模过程中死亡4只,造模成功的24只大鼠随机分为阴虚+薄型组(不予药物治疗)、中药低剂量组(13.65 g·kg^(-1)·d^(-1),灌胃30 d)、中药高剂量组(27.3 g·kg^(-1)·d^(-1),灌胃30 d)、西药组(戊酸雌二醇片0.11 mg·kg^(-1)·d^(-1),灌胃30 d),每组6只。观察6组大鼠子宫内膜组织形态学变化、子宫内膜纤维化、环腺苷酸(Cyclic adenosine monophosphate,cAMP)、环磷酸鸟苷(Cyclic guanosine monophosphate,cGMP)表达变化及miRNA135a、人同源框蛋白A10(Homeobox A10,HOXA10)的mRNA表达变化。结果与正常对照组比较,其余各组的子宫内膜厚度较薄,腺体较少,内膜连续性较差;除正常对照组外,中药高剂量组与其余各组比较,内膜更加完整,腺上皮及腔上皮细胞排列更整齐,腺体和毛细血管也明显增加。与正常对照组比较,肺肾阴虚组和阴虚+薄型组中的纤维化程度、不可溶性胶原纤维含量显著升高;中药高剂量组与其余各组比较,纤维化程度及不可溶性胶原纤维含量降低。与正常对照组比较,其余各组中的cAMP水平显著升高;与阴虚+薄型组比较,其余各组cAMP水平显著下降(P<0.05)。与正常对照组比较,其余各组中的cGMP水平显著降低(P<0.05);与阴虚+薄型组比较,其余各组cGMP水平显著升高(P<0.05)。与正常对照组及肺肾阴虚组比较,其余各组中的miR-135a的mRNA表达水平显著升高(P<0.05);与阴虚+薄型组比较,其余各组的miR-135a含量显著降低(P<0.05)。与正常对照组及肺肾阴虚组比较,其余各组的HOXA10 mRNA�
Objective To explore the effect of Bufei Tianjing Recipe on the receptivity of thin endometrium in rats.Methods Unmated 10 week old SD female with body weight of 180-220 g were selected.After 7 days of adaptive feeding and 10 days of continuous HE staining,40 SD rats with regular estrous cycles were screened.6 female rats were randomly selected as the normal control group,and the other 34 female rats were established as lung-kidney Yin deficiency model.After successful modeling,6 female rats were randomly selected as lung-kidney Yin deficiency group,and the other rats with successfully modeling were established as thin endometrium rat model,4 of which died during the modeling process.The 24 rats were randomly divided into Yin deficiency+thin type group(without medication),low-dose group of traditional Chinese medicine(13.65 g·kg^(-1)·d^(-1),gavage for 30 days),high-dose group of traditional Chinese medicine(27.3 g·kg^(-1)·d^(-1),gavage for 30 days),and Western medicine group(0.11 mg·kg^(-1)·d^(-1),gavage for 30 days),6 rats per group.The endometrial histomorphologic changes,endometrial fibrosis,cyclic adenosine monophosphate(cAMP),cyclic guanosine monophosphate(cGMP),and miRNA135a in 6 groups of rats MRNA expression of human homeobox protein A10(HOXA10)were observed.Results Compared with the normal control group,the other groups had thinner endometrial thickness,fewer glands and poor endometrial continuity.In addition to the normal control group,compared with the other groups,the endometrium was more complete,the glandular epithelium and coelomial epithelium cells were arranged more neatly,and the glands and capillaries were also increased significantly in the high-dose group.Compared with the normal control group,the degree of fibrosis and insoluble collagen fiber content in the lung and kidney Yin deficiency group and the lung and kidney Yin deficiency+thin endometrium rat model group were significantly increased.Compared with other groups,the degree of fibrosis and insoluble collagen fiber content in the
