多西环素联合甲硝唑干预对LPS诱导的大鼠子宫内膜炎Th1/Th2的影响

摘要 目的:探讨多西环素联合甲硝唑对脂多糖(Lipopolysaccharides,LPS)诱导的大鼠子宫内膜炎Th1/Th2的影响。方法:本研究于2022年6月至2022年12月选取无特定病原体(Specific Pathogen Free,SPF)级实验大鼠成功孕育的50只SD雌性大鼠,饲养在福建中医药大学动物实验中心,随机分为空白组、大鼠子宫内膜炎(CE)模型组、多西环素干预组、甲硝唑干预组及联合用药组,每组含10只大鼠,空白组外其余4组由LPS诱导建立大鼠CE模型。连续灌胃给药14d后,取腹主动脉血通过酶联免疫吸附试验(ELISA)检测肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α),白细胞介素-2(Interleukin-2,IL-2)、白细胞介素-4(Interleukin-4,IL-4)、白细胞介素-6(Interleukin-6,IL-6)水平,同时取各组大鼠子宫及附件组织,苏木素-伊红(HE)染色观察各组大鼠组织病理改变情况。结果:模型组较空白组宫腔粘连,宫内膜水肿、血管充血,炎症浸润,联合用药组干预下宫腔粘连、水肿、充血改善显著,无明显内膜损伤。模型组较空白组Th1/Th2水平升高(P<0.05),多西环素组、甲硝唑组及联合用药组Th1/Th2水平较模型组下降(P<0.05),且联合用药组较多西环素、甲硝唑组下降更显著,差异具有统计学意义(P<0.05)。与空白组比较,模型组TNF-α、IL-4、IL-6 mRNA水平降低(P<0.05),IL-2 mRNA水平升高(P<0.05);与模型组相比,联合用药组TNF-α、IL-4、IL-6 mRNA水平降低(P<0.05),IL-2 mRNA水平升高(P<0.05)。结论:多西环素联合甲硝唑干预CE大鼠TNF-α、IL-4、IL-6水平降低,IL-2水平升高,共同调节Th1/Th2失衡,有助于CE的治疗及发展。 Objective:To investigate the effect of doxycycline combined with metronidazole intervention on lipopolysaccharides(LPS)induced rat endometritis Th1/Th2.Method:From June 2022 to December 2022,this study selected 50 SD female rats that were successfully bred without specific pathogen free(SPF)grade experimental rats and raised them at the Animal Experimental Center of Fujian University of Traditional Chinese Medicine.They were randomLy divided into a blank group,a rat endometritis(CE)model group,a doxycycline intervention group,a metronidazole intervention group,and a combination therapy group,each containing 10 rats,Except for the blank group,the remaining four groups were induced by LPS to establish a rat CE model.After continuous intragastric administration for 14 days,tumor necrosis factor(TNF)was detected by enzyme-linked immunosorbent assay(ELISA)using abdominal aortic blood-αTumor Necrosis factor-α,TNF-α,Interleukin-2(IL-2),Interleukin-4(IL-4),and Interleukin-6(IL-6)levels were measured.At the same time,uterine and adnexal tissues of rats in each group were taken and stained with hematoxylin eosin(HE)to observe the pathological changes of the tissues in each group.Result:The model group showed significant improvement in uterine adhesions,endometrial edema,vascular congestion,and inflammatory infiltration compared to the blank group.The combined medication group showed significant improvement in uterine adhesions,edema,and congestion without significant endometrial damage.The model group showed an increase in Th1/Th2 levels compared to the blank group(P<0.05),while the doxycycline group,metronidazole group,and combination group showed a decrease in Th1/Th2 levels compared to the model group(P<0.05).Moreover,the combination group showed a more significant decrease in Th1/Th2 levels compared to the control group(P<0.05),and the difference was statistically significant.Compared with the blank group,the model group TNF-α、The levels of IL-4 and IL-6 mRNA decreased(P<0.05),while the levels of IL-2 mRNA increa
