摘要
目的:探讨补肾调经方对雄激素致不孕大鼠(androgen-induced sterile rats,ASR)子宫转化生长因子β1及其受体(TGFβ1/TGFβRⅠ)表达的影响。方法:给出生9日龄SD雌性大鼠一次性皮下注射丙酸睾丸酮(T),建立ASR模型。设乌鸡白凤口服液为阳性对照药。分为正常组、模型组、阳性药对照组和中药治疗组。ASR模型81日龄始灌服乌鸡白凤口服液、补肾调经方,连续7周,130、131日龄麻醉处死,采用UE染色及免疫组化技术分别观察子宫组织形态学及TGFβ1、TGFβRⅠ表达的变化。结果:补肾调经方可改善病理状态下子宫的形态结构,使模型大鼠子宫内膜、肌层TGFβ1以及肌层TGFβRⅠ的表达增强。结论:补肾调经方可通过提高ASR模型子宫内膜、肌层TGFβ1及肌层TGFβRⅠ的表达,促进子宫的发育,改善子宫内膜的分泌功能,有利于胚泡的植入。
AIM: To observe the effect of Bushen Tiaojing Recipe (Radix remhanniae praeparata, Fructus lycii, Fructus rubi, Herba epimedii, Radix angelicae sinensis, Rhizoma atractylodis macrocephalae, Rhizoma cyperi, Flos carthami, Semen persicae) on expression of transforming growth factor β1 and its receptor (TGFβ1/ TGFβR Ⅰ ) in uterus in androgen-induced sterile rats (ASR). METHODS: The ASR model was established by giving testosterone propionate to SD female rats at the age of 9 d subcutaneous injection. Wujibaifeng Oral Liquid was taken in the positive group. The model rats were divided into Wujibaifeng Oral Lquid group, Bushen Tiaojing Recipe group and medol group, and at the same time, a normal control group of normal rats was also set up. When the ASR were at the age of 81 d, they were fed with Bushen Tiaojing Recipe and Wujibaifeng Oral Liquid for seven weeks. When the rats were at the age of 130 d, or of 131 d, the animals were anaesthetized and killed, The morphological structures of the uterus were observed by HE dyeing. The expression of TGFβ1/TGFβR Ⅰ in the uterus was detected by immunohistochemical method. RESULTS: Bushen Tiaojing Recipe could improve the uterine morphological structures in pathological condition, and strengthen the expression of TGFβ1 in the myometrium and endometrium as well as the expression of TGFβRⅠ in the myometrium in ASR. CONCLUSION: The experimental results suggest that Bushen Tiaojing Recipe may promote growth of uterus, improve secretory function of endometrium through increasing the expression of TGFβ1/TGFβR Ⅰ in the uterus ,which contributed to implantation of blastocyst.
