Argonaute蛋白与心血管疾病关系的研究进展

摘要 Argonaute(AGO)蛋白是一种高度保守的蛋白,可与小分子RNA构成RNA诱导沉默复合物(RISC),而RISC可靶向与小分子RNAs互补的靶信使RNA,并通过RNA降解、翻译抑制或染色质修饰等途径调控基因表达,从而参与生理病理过程。既往关于AGO蛋白的研究主要集中于其在肿瘤疾病发生发展中的作用,近年则着重于AGO蛋白的组织分布特点、复合体结合能力和靶向沉默基因的能力在糖尿病心肌病、心力衰竭、心房颤动等心血管疾病(CVD)发生发展中的作用。AGO蛋白在CVD中的作用机制已成为目前研究的热点,深入探究其具体的分子调节机制有望为CVD的诊疗提供新靶点。 Argonaute(AGO)proteins,a type of highly conserved protein,which can form an RNA-induced silencing complex(RISC)with small RNAs,and RISC can target messenger RNAs complementary to small RNAs and regulate gene expression through RNA degradation,translational inhibition or chromatin modification,thus participating in various physiological and pathological processes.Previous studies focused on the role of AGO proteins in the development of tumor diseases.In recent years,the emphasis has been placed on the roles of the tissue distribution,the capacity for complex binding,and the ability to target silenced genes of AGO proteins in the development of cardiovascular diseases(CVD)such as diabetic cardiomyopathy,heart failure,and atrial fibrillation.The role of AGO protein in CVD has become a major research hotspot,and further exploration of the specific molecular regulatory mechanism is expected to provide new targets for CVD diagnosis and treatment.
