基于中心法则探究碱基突变影响CTPS细胞蛇组装的综合实验

摘要 CTPS细胞蛇是一种无膜细胞器,在细胞中以丝状或环状形式存在.将含有RFP标记的CTPS过表达载体转入Hela细胞中,荧光显微镜下观察红色荧光蛋白形成的丝状或环状结构.通过定点突变关键氨基酸,观察CTPS细胞蛇组装的效果,帮助学生理解分子生物学“中心法则”中DNA碱基序列决定蛋白质一级结构进而决定蛋白质的高级结构,同时激发学生的学习兴趣,培养学生的科学思维,提高综合素质. CTPS cytoophidia are membrane-less organelles that exist as filaments or rings in the cell.CTPS overexpression vectors containing RFP-labeled CTPS were transferred into Hela cells,and the filamentous or annular structures formed by red fluorescent proteins were observed under fluorescence microscopy.Through site-specific mutation of key amino acids,observe the effect of CTPS cytoophidia assembly,help students understand the“central dogma”of molecular biology in which DNA base sequence determines the primary structure of proteins and thus the higher structure of proteins,and cultivate students scientific thinking and improve their integrated quality.
