“新流腺”三联嵌合型病毒样颗粒疫苗的免疫效果评价

摘要 为了评价“新流腺”三联嵌合型病毒样颗粒(ND-AI-FAdV4 cVLPs)的免疫保护效果,本试验将其定量免疫至7日龄SPF雏鸡中,通过血凝抑制试验、ELISA和淋巴细胞增殖检测ND-AI-FAdV4 cVLPs诱导的体液及细胞免疫,并进行攻毒试验评价ND-AI-FAdV4 cVLPs的攻毒保护能力。结果显示,ND-AI-FAdV4 cVLPs可诱导机体产生与NDV弱毒疫苗和AIV灭活疫苗相当的HI抗体效价及针对FAdV4 Fiber2蛋白的高水平特异性抗体,NDV、AIV和FAdV4灭活病毒的分别刺激可诱导ND-AI-FAdV4 cVLPs免疫组脾脏淋巴细胞发生活化及增殖;ND-AI-FAdV4 cVLPs免疫后可提供针对NDV强毒株NA-1株、AIV H9N2株及FAdV4强毒株攻击的有效保护,降低了病毒感染所造成的病理损伤,且缩短了体内病毒载量及体外排毒时间。本研究为ND-AI-FAdV4 cVLPs疫苗的应用奠定了基础,同时也为新城疫、禽流感和禽腺病毒病多联新型疫苗的研发提供了参考。 To evaluate the immunoprotective effect of the triple chimeric virus-like particles(ND-AI-FAdV4 cVLPs),SPF chickens were quantitatively immunized with ND-AI-FAdV4 cVLPs.Hemagglutinative inhibition test,ELISA and lymphocyte proliferation test were performed to detect the humoral and cellular immunity induced by ND-AI-FAdV4 cVLPs,and challenge test was conducted to evaluate the challenge protection ability of ND-AI-FAdV4 cVLPs.The results showed that ND-AI-FAdV4 cVLPs could induce HI-antibody titer equivalent to that of NDV attenuated vaccine or AIV inactivated vaccine and high levels specific antibody against FAdV4 Fiber2 protein.Activation and proliferation of the splenic lymphocytes in the ND-AI-FAdV4 cVLPs immunized group were induced by NDV,AIV and FAdV4 inactivated viruses respectively.In addition,ND-AI-FAdV4 cVLPs could provide effective protection against NDV-strong strains NA-1,AIV H9N2 and FAdV4 strong strains,reduce the pathological damage caused by virus infection,and shorten viral load in vivo and in vitro detoxication time.This study lays a foundation for the application of ND-AI-FAdV4 cVLPs vaccine,and also provides a reference for the development of novel multi-combination vaccines against Newcastle disease,avian influenza and avian adenovirus viruses.
