摘要
采用新城疫病毒样颗粒载体平台和昆虫杆状病毒表达系统,将禽流感H9N2株HA、禽4型腺病毒Fiber2嵌合至新城疫病毒样颗粒载体表面进而构建“新流腺”三联嵌合型病毒样颗粒(ND-AI-FAdV4 cVLPs)。PCR及免疫荧光检测结果显示,成功构建了重组杆状病毒rBV-cFiber2;透射电镜结果显示,ND-AI-FAdV4 cVLPs直径约为100 nm、含有囊膜的空心蛋白颗粒;免疫印迹结果显示,嵌合型病毒样颗粒各组分蛋白均正确表达。本试验为新城疫、禽流感和禽腺病毒病的安全、高效病毒样颗粒新型疫苗候选株的应用奠定了基础。
Based on NDV-like particle vector platform and insect baculovirus expression system,the HA of avian influenza H9 N2 strain and Fiber2 of avian adenovirus type 4 were chimeric on the surface of ND VLPs to construct ND-AI-FAdV4 cVLPs.The results of PCR and immunofluorescence showed that the recombinant baculoviruses rBV-cFiber2 were successfully constructed.Transmission electron microscopy(TEM)showed that ND-AI-FAdV4 cVLPs were encapsulated hollow protein particles with a size of about 100 nm.The results of western blotting showed that the chimeric virus-like particles were all correctly expressed.This study laid the foundation for the application of safe and efficient virus-like particle vaccine candidates against Newcastle disease,avian influenza and avian adenovirus disease.
