摘要
目的:对结肠癌肝转移和肝外卵巢转移的患者进行二代转录组测序,比较转移灶与正常组织RNA表达谱的差异,寻找转移灶的分子差异与作用位点。方法:对1例合并肝与和盆腔转移的回盲部结肠癌患者的结肠上皮与肝、卵巢转移组织进行测序分析,筛选出不同转移灶之间的差异表达基因并进行功能富集分析。结果:与正常组织相比,肝转移灶中差异表达基因12064个,卵巢转移灶中差异表达基因12265个。KEGG功能分析发现转移灶的差异基因主要富集于癌症与PI3K-AKT通路。肝转移灶FZD9、RET、IFNG等基因显著差异表达,卵巢转移灶WNT2、WNT7B、IFNG等基因显著差异表达。结论:转录测序发现该患者转移与正常组织间差异基因主要富集于癌症通路,其中FZD9、IFNG、WNT2等差异表达基因可能与结肠癌转移相关。
Objective:Transcriptome sequencing was performed on an patients suffered from colon cancer with liver and extrahepatic pelvic metastases,and the differences in RNA expression profiles between metastases and normal tissues were compared to find the molecular differences.Methods:A rare ileocecal colon cancer patient with liver and pelvic metastases was sequenced to screen out differentially expressed genes(DEGs)in metastases and functional enrichment analysis was employed.Results:Compared with normal tissue,12064 DEGs were found in liver metastases and 12265 DEGs were found in ovarian metastases.KEGG functional analysis showed that cancer and PI3K-AKT pathways were enriched mostly.FZD9,RET,IFNG were significantly differentially expressed in liver metastases,and WNT2,WNT7B,IFNG were significantly differentially expressed in ovarian metastases.Conclusion:Transcriptome sequencing of the patient found that the DEGs between the metastatic and normal tissues were mainly enriched in pathway in cancer.FZD9,IFNG,WNT2 and other DEGs may be related to tumor metastasis.
