摘要
为探明养殖密度对虹鳟幼鱼生长性能的影响,对高密度(6.99 kg/m^(3))、中密度(4.66 kg/m^(3))、低密度(2.33 kg/m^(3))养殖条件下虹鳟生长和肌肉转录组进行比较分析。试验结果显示:养殖32 d后,低密度组终末体质量等指标均高于中、高密度组;每个样品转录组有效数据均达5.99 Gb,比对效率91.67%~92.77%,共发掘5501个新基因,其中4121个得到注释,1380个未得到注释;低密度组与高密度组、低密度组与中密度组、中密度组与高密度组比较分别获得了2760、654、2316个差异表达基因,其生物学过程和细胞组分均富集在DNA整合、DNA介导的转位和褶皱,分子功能主要富集在细胞骨架的结构组成(低密度组与高密度组、中密度组与高密度组差异表达基因)和DNA结合、氧化还原酶活性作用于成对供体结合或重组(低密度组与中密度组差异表达基因),且分别有920、196、743个差异表达基因定位到相应的212、112、189个特定代谢途径;通过人工筛选获得了19条与生长和应激相关的基因序列。试验结果表明,养殖密度对虹鳟幼鱼生长及相关基因的表达产生显著影响。
In order to study the effect of culture density on the growth of rainbow trout Oncorhynchus mykiss , the growth perfomance and muscle transcriptome of rainbow trout under high density (6.99 kg/m^(3) ), medium density (4.66 kg/m^(3) ) and low density (2.33 kg/m^(3) ) culture conditions were compared and analyzed, respectively. The results showed that after 32 days of breeding, the final quality and other growth-related indexes of low density group were higher than those of medium and high density group. The effective data of For each sample, the effective data of transcriptome all reached 5.99 Gb, and the comparison efficiency ranged from 91.67%—92.77%. Furthermore, 19 gene sequences related to growth and stress were obtained by artificial, screening. The results showed that the culture density had a significant effect on the growth of rainbow trout and the expression of related genes.
