小儿黄龙颗粒对注意力缺陷多动障碍大鼠的干预作用及机制研究

摘要 目的研究小儿黄龙颗粒对注意力缺陷多动障碍(ADHD)模型动物的干预作用及作用机制。方法采用幼龄自发性高血压大鼠(SHR)作为ADHD模型大鼠,将32只健康SHR大鼠按体重随机分为4组,即SHR大鼠组,小儿黄龙颗粒1.88、3.75 g·kg^(-1)组,哌甲酯组,另设WKY大鼠组、Wistar大鼠组,每组8只,连续灌胃给药21 d。以自主活动度、焦虑程度、社交偏好程度、易激惹程度、空间记忆能力、注意定势转移能力作为ADHD行为学指标;以超高效液相色谱-质谱(UPLC-MS/MS)方法检测大鼠大脑内前额叶中多巴胺(DA)、高香草酸(HVA)的含量;以PCR方法检测大鼠前额叶内酪氨酸羟化酶(TH)、多巴胺转运体(DAT)mRNA的表达量。结果3.75 g·kg^(-1)小儿黄龙颗粒可改善ADHD模型大鼠的行为学异常,显著提高大鼠前额叶DA含量(P<0.01)、降低HVA含量(P<0.05),明显促进前额叶TH mRNA、DAT mRNA的表达(P<0.01)。结论小儿黄龙颗粒能改善ADHD模型大鼠的行为学障碍,其作用机制与改善多巴胺能神经功能有关。 Objective To observe the intervention of Xiao’er Huanglong granules on rats with attention deficit hyperactivity disorder(ADHD)and analyze the mechanism.Methods Young spontaneously hypertensive rats(SHR)were used to establish ADHD models.Totally 32 healthy SHR rats were randomized into 4 groups according to the body weight,an SHR group,Xiao’er Huanglong granule 1.88 and 3.75 g·kg^(-1) groups and a methylphenidate group.while a WKY group and Wistar group were also set up,with 8 rats in each group.The rats were given Xiao’er Huanglong granules by gavage for 21 days.The degree of autonomous activity,anxiety,social preference,irritability,spatial memory ability and attention set transfer ability were used as the behavioral indicators of ADHD.The levels of dopamine(DA)and homovanillic acid(HVA)in the prefrontal cortex of the rats were detected by ultra-high performance liquid chromatography-mass spectrometry(UPLC-MS/MS).The expressions of tyrosine hydroxylase(TH)and dopamine transporter(DAT)mRNA in the prefrontal cortex of the rats were detected by PCR.Results Xiao’er Huanglong granules(3.75 g·kg^(-1))significantly improved the behavioral abnormalities of ADHD model rats,significantly increased the level of DA(P<0.01),reduced the level of HVA(P<0.05),and promoted the expression of TH mRNA and DAT mRNA in the prefrontal cortex(P<0.01).Conclusion Xiao’er Huanglong granules can improve the behavioral disorder in ADHD model rats,whose mechanism is related to the improvement of dopaminergic neurological function.
